It was the first time to utilize the suspension cultured-cells of Jasminumamplexicaule to transform dihydro-epi-deoxyarteannuin B (1). Through severalextraction and purification methods, two products were obtained, they wereelucidated by physico-chemical properties and spectra methods as follows:7α-hydroxy-dihydro-epi-deoxyarteannuin B (2) and3α-hydroxy-dihydro-epi-deoxyarteannuin B (3), among them, product2was a new compound.Time-course curve was established and the results showed that the conversion rate ofproduct2was up to maximum29.02%in the first day, while product3was23.26%also appeared in the same day. Substrate1had been showed to be one of the mostimportant biosynthesis intermediates of biosynthetic pathway of artemisinin and hadbeen studied a lot about its pharmacological effects. However, their biosyntheticpathway has not yet been elucidated clearly. Therefore, it was quite significant toexplore their intermediates among the biosynthesis pathway.Meanwhile, we selected transgenic hairy roots of Fallopia multiflora tobiotransform artemisinic acid (4). The products had been detected by HPLC andthrough several extraction and purification methods, two products were obtained byusing several chromatography methods. Their structures were elucidated as3-β-hydroxyartemisinic acid (5) and arteannuin B (6). Time-course curve wasestablished in order to make sure the best conversion time. The comparison ofproducts of hairy roots of F. multiflora and the suspension cultured-cells of J.amplexicaule also detected by HPLC in order to further investigate transformationrelations of the various intermediates in the biosynthetic pathway of artemisinin.In addition, hydrogen peroxide and salicylic acid as the two most commonexogenous factors were added into the control groups to test their impacts on thetransformation. The results showed that were not conducive to generate the products.Products2and3were detected by inhibition of tumor cell line K562and HeLa, theresults showed that both of them had the antitumor effect, and that of product2wasbetter than product3.
