Objectives:To observe the influence of perfluorocarbon emulsions, UW solution, and themixed solution of them two on antiinflammation, antioxidation, histopathological andultrastructural of isolated rat lung at different time point of preservation. The degreeof oxidative damage, the expression of inflammatory factors, histopathologicalchanges and ultrastructural changes of pulmonary were examined to evaluate theprotective effects of three kinds of solution. The aim of study was to search for aneffective and safety way to store donor lungs.Methods:The72SD rats were divided into six groups randomly, and12in each group. Theisolated lung perfusion models were constructed on all the rats, then took out thewhole of heart and lungs,stored in lung preservation solution. UW-6and UW-12groups were flushed and stored with UW solution, and stored for6and12hoursrespectively. UW/FCE-6and UW/FCE-12groups were flushed and stored with themixed solution of UW solution and FCE, and the store time as above. FCE-6andFCE-12groups were flushed and stored with FCE, and the store time as above.Specimens were harvested and the degree of oxidative damage, the level ofinflammatory factors, the histopathological changes, ultrastructural changes andpathological score of pulmonary tissue were analyzed.Results:(1) The IL-1β、IL-6and TNF-α were no significant differences among threesolutions (P>0.05);(2) The IL-1β、IL-6and TNF-α were no significant differencesbetween6hours and12hours (P>0.05);(3) MDA、MPO and pathological score of6hours were significantly lower than of12hours (P<0.05), SOD of6hours weresignificantly higher than of12hours (P<0.05);(4) SOD of FCE groups weresignificantly higher than of UW/FCE groups (P<0.05), MDA of UW/FCE groupswere significantly higher than of the other two groups (P<0.05), MPO and pathological score of FCE groups were significantly lower than of the other twogroups (P<0.05);(5) The ultrastructural changes and histopathological changes inpulmonary tissue were seriously with UW/FCE and UW but lightly with FCE.Conclusion:Compared with UW solution, perfluororcarbon emulsions can also improveantioxidant ability of lung, but the mixed solution of FCE and UW solution were theworst on the antioxidant ability of lung; FCE were much better to inhibit the assemblof inflammatory cells, they can abate and delay lung injury much better during storage.We consider that, useing perfluororcarbon emulsion as preservation solution alone is agood choice.
