Aim: To investigate the expression of CDX2and Villin proteins in gastriccardiac adenocarcinoma (GCA) and different grade lesions of gastric cardiac tissueand subtypes of cardiac intestinal metaplasia (IM), simultaneously evaluate therelationship of these protein expressions and carcinogenesis of gastric cardiaccarcinoma and intestinal metaplasia of gastric cardiac.Method: The expressions of CDX2and Villin proteins were evaluated byimmunohistochemistry in25cases of cardiac gastritis tissues with IM,48cases ofatypical hyperplasia tissues with IM,55cases of gastric cardiac adenocarcinomatissues and15cases of normal gastric cardiac tissues, which were obtained by gastriccardiac biopsy or surgical resection from oncology department of local hospitals inHenan province,China. Mucus staining was used to subtype the73tissues of gastriccardiac IM and analyzed the correlation between the expression and the subtypes ofcardiac IM by SPSS soft.Result:1.The expressions of CDX2and Villin proteins in normal tissues,intestinal metaplasia tissues, atypical hyperplasia tissues and gastric cardiac cancertissues were difference and increased with staging (P<0.05). No CDX2and Villinexpression was found in normal gastric cardiac tissues. The positive rates of CDX2protein in cardiac gastritis with IM, dysplasia with IM, and carcinoma tissues were80.0%(20/25),62.5%(30/48) and34.5%(19/55),respectively. While the positive ratesof Villin protein in cardiac gastritis with IM, dysplasia with IM, and carcinoma tissueswere76.0%(19/25),60.4%(29/48) and36.4%(20/55), respectively. The result showinga significant difference among the three group both for CDX2and Villin (P<0.01).2.The positive rates of CDX2protein and Villin protein in subtypes I IM were88.0%(22/25) and92.0%(23/25), in subtype II IM were76.2%(16/21) and71.4%(15/21), in subtype III IM were44.4%(12/27) and40.7%(11/27) respectively. Chi-square analysis showed the expression leve were significant different among threesubtypes both for CDX2protein and Villin protein.3. Either the expression of CDX2or Villin was positively correlated with tumor differentiation (both P<0.05), but wasnot associated with age, sex, invasion and metastasis of lymph node (P>0.05).Conclusion:1.CDX2and Villin proteins may play an important role in theoccurrence of gastric cardiac cancer and maybe protect factors.2. Both of CDX2andVillin proteins was detected in the earlier stage of gastric cardiac IM and theirexpression was related with subtypes of gastric cardiac IM. Subtype III IM mayclosely relate with the occurrence of gastric cardiac cancer.3. CDX2and Villinproteins are helpful to judge the degree of malignancy of GCA.
