ObjectiveApplication of loop-mediated isothermal amplification (LAMP) method to pneumoniapatients sputum about common pathogenic bacteria of nucleic acid test, at the sametime and Application of sputum cultures. comparison the two method, this paperdiscusses the value of LAMP method in the lower respiratory infections patients, Thebeginning starts a treatment has covered pathogens group and treated first not coverpathogens group of two groups of patients clinical condition, Prove the importance ofthe rapid cause examinationMethodsCollect diagnosis of fujian province qualified sputum samples in289cases whichwere lower respiratory tract infections Pathogen were detected from the sputumsamples by the routine bacterial culture and loop-mediated isothermal amplificationtechnique. To analyze the distribution of pathogenic bacteria causing lowerrespiratory tract infections in respiratory department, and at the same time test thesensitivity and specificity of the the system; compare the result of the routine bacterialculture and loop-mediated isothermal amplification technique. The test results>1x103cases are divided into groups: treated first has covered pathogens and treated firstnot cover of pathogens, compare two groups of age, patient basic diseases,anti-infection treatment time, the whole hospital days, The beginning starts of cureefficient and cure rate.Results1. The test system has high sensitivity and specificity, among289cases,44casebacteria were isolated with the routine bacterial culture, the bacterial positive rate was15.2%. with the LAMP method in detecting bacterial concentrations>1x103, judgeas a positive result of the124cases, the bacterial positive rate was43.0%, Had significant differences compare with the the routine bacterial culture (P <0.05);2.arialyze the distribution of pathogenic bacteria of the lower respiratory infectionspatients, the distribution of bacteria accounted for50.8%, The non-typical pathogenicbacteria accounted for14.2%; Gram-negative bacteria as the main pathogenic bacteria, the top four were pseudomonas aeruginosa40strains, haemophilus influenzae34strains, Klebsiella pneumoniae20strains, streptococcus pneumoniae20strains. Thenon-typical pathogenic bacteria41strains pathogens, including pneumoniamycoplasma39strains, pneumonia chlamydia2strains.3. beginning starts atreatment has covered pathogens group and treated first not cover pathogens groupHad significant differences of the the whole hospital days and the beginning starts tocure efficient (P <0.05); But more anti-infection treatment days and cure rate, bothanti-infection treatment time and the whole cute rate were not statistically significant(P=0.960).ConclusionsWith LAMP method can rapidly Rapid detection of the bacteria in the sputum, and thepositive rate is high compare with the routine sputum cultures. When the bacteriaconcentration>1x103, the positive rate was43.0%,the sensitivity is74.0%, thespecificity is95.3%.The LAMP method has the higher sensitivity than the routinesputum cultures. Before treatment understand the pathogenic bacteria, can shortenhospital days and improve the The beginning starts of cure efficient.
