Objective To establish acute pancreatitis(AP) animal model in rats, dynamicallydetect serum trypsinogen activation peptide(TAP) and C-reactive protein(CRP) ofacute pancreatitis in rats. To investigate the role of serum TAP and CRP in occurrenceand development of AP. It can provide a theory basis for AP early diagnosis.Methods96specific pathogen free(SPF) Wistar male rats were divided into twogroups randomly (n=48in each group): sham-operated group and AP model group.sham-operated group only flip open the abdominal cavity and touch pancreas severaltimes, AP model group was induced by injection of5%Na-Tc under the membrane ofpancreas to establish AP animal model. The rats were killed at3rd,6th,12thand24thhour after modeling in every group. A portion of pancreatic tissues were observed byhematoxylin-eosin(HE) staining. The concentration of amylase in serum was measuredby holo-automatic biochemical analyzer. We detected the concentration of TAP andCRP in serum with enzyme-linked immunosorbent assay(ELISA) method.Results We successfully established acute pancreatitis animal model. The rats ofpancreatitis group exhibited high amylase and pancreatic pathological injury aftermodeling. The pancreatic histopathologic scores in sham-operated group compared APgroup at3rdhour was no significant difference (P>0.05). The pancreatichistopathologic scores in sham-operated group at6th,12th,24thhour with AP group atthe same time and showed significant difference (P<0.05). The pancreatichistopathologic scores in sham-operated group had no significant differences (P>0.05)compared with each other. The pancreatic histopathologic scores in AP group when showed significant differences (P<0.05) compared with each other. The concentrationof TAP in sham-operated group compared AP group at the same time was significantdifference (P<0.05). The concentration of TAP in sham-operated group had nosignificant differences (P>0.05) compared with each other. The concentration of TAPin sham-operated group has significant differences (P<0.05) compared with each other.The concentration of CRP in sham-operated group compared AP group at3rdhour wasno significant difference (P>0.05). The concentration of CRP in sham-operated groupat6th,12th,24thhour compared AP group at the same time was significant difference(P<0.05). The concentration of CRP in sham-operated group at3rdhour compared with6thhour, and12thhour compared with24thhour has no significant differences (P>0.05),6thhour compared with12thand24thhour had significant differences (P<0.05). Theconcentration of CRP in AP group has significant differences (P<0.05) compared witheach other. The concentration of TAP and CRP showed higher positive correlation withthe pancreatic histopathologic scores.Conclusion Serum TAP can be used to diagnosis of AP in early stage for judgment.TAP and CRP detected dynamically in AP in early stage can be used as indicators ofserverity, which plays an important role in forecasting AP.
