Object: To explore the relationship between endothelin-1gene polymorphisms inlocus rs5370, rs1630736and the morbidity and progress of Henoch-Schonleinpurpura, the relationship between the plasma, urine levels of endothelin-1and themorbidity and progress of HSP, the relationship between the plasma, urine levels ofendothelin-1and the ET-1gene polymorphisms,to further explore the pathogenesisof this disease and provide diagnosis, treatment and prognosis of Henoch-Schonleinpurpura with possible scientific basis.Method: Thirty seven children with HSP are enrolled as case group,including18children with HSP and19children with Henoch-Schonlein purpura nephritis.100healthy children are enrolled as gene control group, of which69children are enrolledas control group of plasma level of endothelin-1.In addition, twenty cure pnumoniainpatient are enrolled as control group of urine level of endothelin-1.Using means ofpolymerase chain reaction and gene sequencing to detect gene polymorphisms ofendothelin-1in locus rs5370and rs1630736. And test the levels of endothelin-1inplasma and urine using radioimmumoassay.Results:(1)In locus rs5370, the differences between case group and controlgroup,HSP group and control group, HSPN group and control group on genefrequencies and allele frequency distribution have no statistical significance (P>0.05).(2) In locus rs1630736, the differences between case group and controlgroup,HSP group and control group, HSPN group and control group on genefrequencies and allele frequency distribution have no statistical significance (P>0.05).(3)In combined genotype, the difference between case group and control group ongene frequencies distribution has no statistical significance(P>0.05).(4) The plasmalevel of ET-1in case group of acute phase is significantly higher than recovery phase(t=2.64,P=0.012) and control group(t=2.12,P=0.042).(5) The differences of urinelevel of ET-1between case group and control group(t=3.52,P=0.001),HSPN groupand control group(t=3.16,P=0.006),the acute phase and the recovery phase of case group(t=2.41,P=0.026)all have statistical significance.(6)In locus rs5370, thedifferences among GG genotype, GT genotype and TT genotype in case group onplasma level of ET-1have no statistical significance (P>0.05),the differencebetween GG genotype and GT genotype in case group on urine level of ET-1has nostatistical significance (P>0.05).(7) In locus rs1630736, the differences among CCgenotype, CT genotype and TT genotype in case group on plasma and urine level ofET-1have no statistical significance (P>0.05).Conclusion:(1) It can not be proved that the endothelin-1gene polymorphisms inlocus rs5370and rs1630736have relationship with morbidity and renal lesson ofHenoch-Schonlein purpura.(2) The plasma and urine level of ET-1can be testedto monitor the progress of the disease in clinic.
