Haloxylon ammodendron (C.A.Mey) Bunge is an imporant desert plant in northwest of China, to its breeding ways, people have been giving more and more attention. So far, the three main reproductive ways of H. ammodendron are seed breeding, branch cuttage and tissue culture, etc. The seed breeding,begining with seed germination,is a major way of h. ammodendron propagation under natural conditions. So seed germination is obviously important. And the tissue culture technology can be used as the preferred way of h. Ammodendron rapid micropropagation.The seeds germination and tissue culture of Minqin H. ammodendron are studied preliminary. Finally,it is found that:The earliest germination time of Minqin H. ammodendron seeds is5h, with high germination rate and vigor.Seeds germination can adapt wide temperature cycle,4℃/17℃,4℃/27℃,17℃/27℃and the constant temperature24℃,with high germination rate and vigor. The germination rate is over80%by treating the H. ammodendron seeds in0.05mol/L-0.4mol/L NaCl, and the germination rate and index decrease sharply when NaCl concentration is higher than0.6mol/L, and drop to zero at1.2mol/L.Although, it would have a certain recovery germination rate under the treatment without NaCl. However, higher concentrations made some seeds losing germination ability permanently.The preferred explant for callus induction is hypocotyl rather than cotyledon.lt is found that: MS (pH5.8)+Agar powder (1%)+sucrose (3%)+2,4-D (1,2,3mg/L) alone and the medium with several certain concentrations of6-BA can both induce callus successfully. MS (pH5.8)+Agar powder (1%)+sucrose (3%)+2,4-D(2.0mg/L)+6-BA (0.5mg/L) was the best for callus inducing. Through the adventitious bud and adventitious roots come from calli, the best medium respectively was:MS (pH5.8)+Agar powder (1%)+sucrose (3%)+6-BA (5.0mg/L)+NAA(1.0mg/L)+2,4-D(0.5mg/L), MS (pH5.8)+Agar powder (1%)+sucrose (3%)+IAA(5.0mg/L)+6-BA(0.2mg/L)+NAA(1.0mg/L).The shoot and root regeneration rate are respectively up to66.3%and80.9%.The best explant for shoot and root regeneration are cotyledon and hypocotyl, respectively.
