Eucommia ulmoides Oliv. is a famous Chinese endemic medicinal tree, and is also one high quality resource of natural rubber. It is rich of Terpenoid such as iridoids and polyterpene, and these active components have attached great attention to the pharmaceutical and industrial industry. The iridoid has been proven to have evident function of anti-cancer, anti-aging, antihypertensive, hypolipidemic, and enhancing immune function, which are widely used in medicine, health, food, and get more and more attention at home and abroad. Eucommia ulmoides gum has an unique duality characteristic of rubber-plastic, and it is far beyond the para rubber in plasticity, thermoelastic, blending, high insulation and corrosion resistance in the low temperature, and it can be widely used in some fields such as rubber industry, aerospace, military industry, shipbuilding, chemical industry, medical. However, the mechanism of terpenoids material synthesis and accumulation of molecular is unclear. Methyl valerate (MVA) pathway is one of the important ways for synthesis of terpenoids compounds.This study was based on data of the transcriptome sequencing of eucommia fruit and leaf, and analysis, the difference of gene expression, sequence characteristics and structural characteristics of Methyl valerate (MVA) pathway, were completed in detail by using bioinformatics methods. The study provided a theoretical basis for the molecular mechanism of the MVA pathway of terpene biosynthesis, genetic mechanisms of efficient accumulation and molecular breeding research.MVA pathway is one important way of terpenoids synthesis. From the transcriptome data of young fruit and mature fruit of eucommia, MVA pathway has been annotated30Unigene, and we found that acetyl-CoA C-acetyltransferase, hydroxymethylglutaryl-CoA synthase, hydroxymethylglutaryl-CoA reductase, mevalonate kinase, phosphomevalonate kinase, and diphosphomevalonate decarboxylase were respectively annotated7,3,11,2,2,5. Differences analysis of gene expression showed that the genes, EuACOT2—EuACOT6、EuHMGS1—EuHMGS3、EuHMGR1、EuHMGR2、EuHMGR8、EuHMGR10、 EuMK1、 EuMK2、EuPMK1、EuMDP1—EuMDP5, were differentially expressed genes for the young fruit and mature fruit. And the genes, EuHMGR4and EuHMGR5, specifically expressed in young fruit; EuACOT7, EuHMGR10specifically expressed in mature fruit.The study identified3full-length cDNA in MVA pathway of young fruit and mature fruit. Full-length cDNA of EuACOT1gene has1248bp, and coded416aa, being stable hydrophobic protein; that of EuHMGS2gene has1356bp, and coded452aa, being the unstable hydrophobic proteins. And full-length cDNA of EuHMGR9gene has1770bp, and coded590aa, being unstable hydrophobic proteins. The secondary structures of above three proteins are predominantly hybrid structure of alpha helical and spiral ring structure.From the transcriptome data of young fruit and leaves of eucommia, MVA pathway has been annotated23Unigene, and we found that acetyl-CoA C-acetyltransferase, hydroxymethylglutaryl-CoA synthase, hydroxymethylglutaryl-CoA reductase, mevalonate kinase, phosphomevalonate kinase, and diphosphomevalonate decarboxylase were respectively annotated3,3,8,2,1,6. And the genes, EuACOT8, EuACOT10, EuHMGS4—EuHMGS6, EuHMGR17, EuHMGR18, EuMK3, EuMK4, and EuMDP7—EuMDP11, specifically expressed in young fruit.The study identified8full-length cDNA in MVA pathway of young fruit and leaves. Full-length cDNA of EuACOT8、EuACOT9、EuACOT10、EuMK3were respectively1224bp、1242bp、1224bp、990bp, and at the same times respectively coded408、414、408、330aa, being stable hydrophobic protein. That of the gene of EuHMGR19was1740bp, coding590aa, and these were hydrophilic protein. Full-length cDNA of the gene of EuHMGS4and EuHMGR18were respectively452,588aa, being unstable hydrophilic protein. The secondary structure of above eight protein are predominantly hybrid structure of alpha helical and spiral ring structure.
