Wheat is one of China’s major grain crops, especially of the staple food of the people of theNorth, and it`s also an important national food reserves. Shandong is one of the biggist wheatgrowing province. Research and development of wheat production has very vital significance, tomeet the food needs of the people, improve the level of urban and rural residents’ material lifeand promote the development of national economy. In this paper,41unique wheat cultivars ofShandong province were collected to construct DNA fingerprint database by SSR technique. Theprincipal results of the experiment are as follows:1.46pairs of SSR primers screened from20amplified with type clear and stable coreprimers. The results showed that polymorphic varance was observed in20pairs of midum and69equipotential variance was detected. The number of equipotential variance of each SSR loca is1to8, and the average is3.45. The fragments size of69polymorphic is between104bp to447bp.And polymorphic information coefficient is0.1695to0.9525and the average is0.703. Thus,there is a big difference in the type and distribution of allelic variants, indicating that the20pairsof core primers showed good polymorphism in the germplasms.2. The20pairs of core primers filterred clear positioning of the20loci, located at the A, B,D three genome.20loci of the distribution is uneven,5,5, and10respectively. Among the69equipotential variance, the number of equipotential variance of genome D is the most (32), nextgenome B (19), and genome A is the least (18). The richness degree of equipotential variance ofGenome A, B and D is3.60,3.80and3.20, respectively, and their genetic diversity index is0.694,0.770and0.675, respectively. Therefore, a comprehensive analysis of the average geneticrichness and genetic diversity index, two indicators, shows that genome B has the highest geneticdiversity, while A and D genome diversity index decreased significantly.3. Based on the principle of belt type clear and repeatable, obtained41wheat materials ofDNA fingerprint database, by data records for69alleles. Genetic differences from the database,you can see that the species, the DNA fingerprint difference is larger, easier to identify. Such asYouzimai and Jimai20, there are18pairs of primers can be separated. Small genetic differencesof wheat varieties, although their DNA fingerprints differences relatively small, but still easy to be identified, including half, and half-brother to the hybrids with same parents. Such as Jimai2and Jimai4, Jimai20and Liangxing99.4. Through UPGMA cluster analysis using NTSYS, it was found that41kinds of materialscould be divided into three groups with the threshold at genetic similarity of0.68. Theexperimental results show that, the varieties promoted in recent years, together ratio increase,suggests that they have strong convergence. Some offspring with the parental relationship can beshowed by the dendrogram. The genetic similarity coefficient of varieties, who has the sameparent, is higher than average, but not the highest value. Illustrates that even if the same breedingparent, generations of genetic differences will also bigger, which breed traits difference. Alsosuggests that these genetic differences of the parents, applies to selective breeding, at the sametime.This study indicated that SSR technique could effectively detect the genetic relationshipsamong the various wheat cultivars, and it could meet the requirements for the construction ofwheat DNA fingerprint database and provide theory basis for collecting, storing, classifying,evaluating, and establishment of the core germplasm of wheat cultivars genetic resources inShandong province.
