Dendrobium, the second largest genus of the orchid, has a high appreciated and medicinal value, and plays an important role in the flower and medicinal market. As Dendrobium demands high quality living environment and achieves a low ability to reproduce, it can’t satisfy the fast-growing demand. Moreover, the long-term over-exploitation not only causes an extreme scarcity of resources, but also brings enormous wild species on the edge of extinction. Therefore, the research and conversation of allows of no delay. We need an effective technical means, which has a function in protecting and utilizing resources, to develop Dendrobium continuously.The fast breeding and germplasm preservation techniques of Dendrobium become a key way to meet market demand and protect endangered species. Taking Dendrobium nobile and Dendrobium candidum as the objects, this study establishes a suit of technology, which uses Dendrobium adult plant tissue to induce rapid propagation of protocorm, and an effective protocorm cryopreservation technique. In this way, a set of system to develop, utilize and protect Dendrobium sustainably is built up.Major achievements1. The establishment of an efficient technique to induce Dendrobium protocormBy inducing culture on apical shoot tips, stems segment of Dendrobium organizations, this study ensures that selecting Dendrobium shoot tips as inducing material can reach a better induction efficiency rate. By orthogonal experiment, which chooses MS, B5, N6as basic culture medium and naphthylacetic acid, and6-BA,2,4-D as Hormone, this paper confirmes that Dendrobium shoot tips is the best induction medium for Dendrobium protocorm. On this basis, we established a complete set of Dendrobium protocorm induction technology.2. The establishment of a cryopreservation technique for Dendrobium protocormWe perform the experiment on protocorm, maintain its free water content decreasing through a multi-step dehydration process, and cryopreservate it under protection. This study cryopreserves Dendrobium protocorm by embedding method, vitrification method and encapsulation-vitrification Method, and finally finds the third way is the best one and proper for protocorm cryopreservation.
