Latex is some special substance that is exuded from plant latex tube. Mulberry latex mainly concents protein, sugar, fats, and alkaloids. Mulberry contains alkaloids1-deoxynojirimycin (DNJ) which is a kind of α-glycosidase inhibitor. Although DNJ mainly exists in leaf、stem and root of mulberry tree, a certain DNJ is found in seeds of mulberry tree. So mulberry latex has important research value. Most researchers put their focus on DNJ of root、stem、leaf and bark of mulberry tree that grown tree, but only a few study measured DNJ content of mulberry sapling and seeds. This paper mainly study the extraction and content determination the accumulation of DNJ from the seeds to mulberry sapling and Mulberry resistance to three kinds of aphid.This will lay the foundation for mulberry latex formation rule and Mulberry resistance to insect.The seeds of mulberry were soaked into warm water with the temperature was40℃,and were cultivated in illuminationincubator with the temperature was28~32℃. Separated their roots、stem and leaves and put them in normal temperature until they were dry.A Reverse-phase high-performance liquid chromatography with UV detection (RP-HPLC-UV) method for the determination of DNJ content in the leaves, stem,roots of mulberry sapling, berry, activaty seed, germinate seed after Pre-column derivatization with9-methyl chloride methyl fluorine (FMOC-Cl) was developed. DNJ was extracted with65%ethanol, derived with FMOC-Cl in borate Potassium buffer solution (pH=8.5) and analyzed by RP-HPLC-UV.The DNJ-FMOC produced could absorb ultraviolet radiation and then was tested with ultraviolet radiation inspector. The results showed that the separation chromatogram of the deravation production was very satisfactory. The peak area of DNJ-FMOC was corresponding with the concentration of DNJ.The method for the determination of DNJ could be used as an effective method in determining the DNJ content in organizations of mulberry and related drugs.The content of DNJ of roots, stems, leaves of mulberry sapling, berry, activaty seed, germinate seed determined by Reverse-phase high-performance liquid chromatography with UV detection using65%ethanol as solvent extraction indicated that the content of DNJ was different in different parts of mulberry tree. In the seeds the DNJ content was higher.The DNJ content was lower along with the seeds growth. Then added and steady gradually.The contents of DNJ of the leaves and stem determine d by Reverse-phase high-performance liquid chromatography with UV detection using65%ethanol as solvent extraction are0.3406%and0.19224%,and the contents of DNJ of the leaves and stem using0.05mol/l hydrochloric acid as solvent extraction are0.16228%and0.09612%.This result indicated that the content of DNJ of the leaves and stem determined Reverse-phase high-performance liquid chromatography with UV detection using65%ethanol is higher than that using0.05mol/l hydrochloric acid as solvent extraction.Using free-hand section and paraffin method observed mulberry structure and the distribution of laticifer, the results showed that the laticifer of mulberry is nonarticulated laticifer. They mainly distributed in cortex and phloem of roots, stems and leaves of mulberry tree and in various organs have difference distributed number.By suction method respectively, leaf immersion method, dip bug method to determine the resistance to aphids of different concentrations of mulberry leaf and latex. We found that the dead quantity of three kinds aphid are most in20%latex of mulberry latex no matter which method was used. The results indicated that latex of mulberry leaves and latex had strong killing action. When the concentration of mulberry leaves were same to latex and the dealing time were same, the toxicity was obviously higher than mulberry leaves. To a certain extent, the processing time was longer, the mortality rate of aphid were more and more obviously with the dealing time extending.
