Latex is some special substance that is exuded from plant latex tube and is widely found among most maraca plant species. Mulberry (Moraceae) latex mainly contains protein, sugar, fats, and alkaloids. Mulberry contains alkaloids 1-deoxynojirimycin (DNJ) which is a kind ofα-glycosidase inhibitor. 1-deoxynojirimycin (DNJ) as natural products of plant products is efficient treatment of diabetes. The DNJ content of Mulberry latex surpass greatly their organizations of mulberry, so mulberry latex has important research value .The paper make some analysis about the extraction and content determination of DNJ in Mulberry latex, as well as the Hypoglycemic effect of mulberry latex. The Study provides a theoretical reference for the comprehensive utilization of mulberry latex.A Reverse-phase high-performance liquid chromatography with UV detection (RP-HPLC-UV) method for the determination of DNJ content in mulberry latex after Pre-column derivatization with 9-methyl chloride methyl fluorine (FMOC-Cl) was developed. Mulberry latex was extracted with 70% ethanol, derived with FMOC-Cl in borate Potassium buffer solution (pH=8.5) and analyzed by RP-HPLC-UV.The DNJ-FMOC produced could absorb ultraviolet radiation and then was tested with ultraviolet radiation inspector. The results showed that the separation chromatogram of the deravation production is very satisfactory. The peak area of DNJ-FMOC was corresponding with the concentration of DNJ. The method for the determination of DNJ of mulberry latex by RP-HPLC-UV which is high recovery, good reproducibility, high precision, and good stability showed that the method was credible and it could be used as an effective method in determining the DNJ content in mulberry latex, other organizations of mulberry, and related drugs.The 1-deoxynojirimycin (DNJ) contents of mulberry latex in different parts of mulberry were investigated in the paper by RP-HPLC-UV. The results showed that there were quite DNJ content changes (wet weight) in latex of different parts of mulberry. The highest DNJ content in Mulberry latex exists in the middle part of mulberry branches.The average is 0.63376%.The lower DNJ content in mulberry latex exists in the peak of Mulberry tress. The average is 0.58128%. The lowest DNJ content in Mulberry latex exists in the base of Mulberry trunk. The average is 0.16382%.The 1-deoxynojirimycin(DNJ) contents(wet weight) in latex of the middle part of mulberry Branches of four mulberry varieties included"Husang 32","Nongsang 8","Nongsang 14", and"Yu 71-1".The results showed that there were some differences about DNJ contents of mulberry latex in four varieties of mulberry. The highest DNJ content in mulberry latex exists in"Nongsang 14". The average is 0.41312%.The DNJ contents of mulberry latex in other three varieties of mulberry were 0.15861%(Husang 32),0.27855%(Nongsang 8),0.27962%(Yu 71-1).In different seasons, the mulberry latex was collected from three different parts of"Nongsang 14"including the peak of tress, middle branches, and the base of trunk. The DNJ contents (wet weight) after purification were determined. The results showed that in all four seasons the highest DNJ content in mulberry latex existed in the middle part of mulberry branches, and the lowest DNJ content in mulberry latex existed in the base of mulberry trunk. In different four seasons, the DNJ contents in latex of the middle part of mulberry branches were quite different. The DNJ contents respectively were 0.10352% in spring, 0.63376% in summer, 0.41312% in autumn and 0.13588% in winter. The highest DNJ content of latex existed in autumn. The DNJ contents in latex of the base of mulberry trunk respectively were 0.04836% in spring, 0.16382% in autumn, 0.16459% in summer, and 0.06602% in winter.The high blood sugar mice were fed with mulberry latex by 2.0496 mg/d, 4.0992mg/d respectively. Compare the decrease percentage of blood glucose levels of different groups of mice including A group of mulberry latex group of mulberry latex and positive control group. After seven days, the blood-sugar value of the different groups high blood sugar mice decreased by 22.22 % ,6.43 % ,26.8 % respectively. After fourteen days, the blood-sugar value of the different groups high blood sugar mice decreased by 26.06%,12.27,21.92%respectively. The results showed that mulberry latex on hyperglycemia mice induced by the streptozotocin(STZ) have significant hypoglycemic effect. After seven days, the hypoglycemic effect of mulberry latex was close to positive control group (Tcarbose Tablets). After fourteen days, the hypoglycemic effect of mulberry latex exceeded positive control group (Tcarbose Tablets).Extract, mulberry latex could inhibited weight loss of diabetic mice. At the same time, mulberry latex also has inhibitory effects on the increase of food intake and water intake about diabetic mice.The experiment results about the determinations of DNJ content in mulberry latex provide theoretical basis for exploring the distribution of DNJ in mulberry tissue, as well the collection of mulberry latex. The results of hypoglycemic effect of mulberry latex provide a research direction for further exploiting diabetes drugs and health food.
