IMF is mainly present in the epimysium and perimysium, and has a medium to high heritability of important economic traits. And it is a main precursor for the formation of muscle flavor, because it affects meat tenderness, flavor and juiciness, especially the meat juicy. H-FABP and L-FABP belong to the FABPs family members, the main function is to promote the intracellular transport of long chain fatty acids, to assist in the fatty acid transport to the place of the β-oxidation, and glycerol parts of the three ester and phospholipid synthesis. In the process of the lipid metabolism, H-FABP and L-FABP have an important role in the uptake and transporter of fatty acids. LPL belongs to the lipase gene family, is the hydrolysis enzyme to catalytic protein-linked triglyceride, plays a central role in the control of fat deposition, controls the triglyceride distribution of fat and muscle, especially in the intramuscular fat synthesis. Therefore, study H-FABP, L-FABP and LPL gene has a great significance to the formation of intramuscular fat.In this study, we used RT-PCR combined with cloning and sequencing methods to get the full sequences of H-FABP gene and L-FABP gene of tianfu goat, which was submitted to the NCBI, for two gene accession number (HQ404383, and HQ606485). Nucleotide homology analysis showed that:tianfu goat H-FABP gene and L-FABP gene had a high homology with other species (mammals). The amino acid comparison showed that:the results of the two proteins were highly conserved. Bioinformatics analysis showed that the phosphorylation sites projections indicated that H-FABP amino acid sequence contained eight phosphorylation sites were2serine,5threonine and a tyrosine sites, and L-FABP amino acid sequence contained two phosphorylation sites were serine sites; most of the H-FABP protein and L-FABP protein had the hydrophilic amino acid residues; secondary structure prediction showed the H-FABP and L-FABP amino acid sequence were composed of α-helix, random coil and extended fragment, and the main was coil; both had kept a lipocalin superfamily.Real-time fluorescent quantitative RT-PCR technology analysis found that H-FABP, L-FABP and LPL gene were detected in the selected12-month-old Tianfu goat of six tissue organization. H-FABP gene had a high expression level in heart tissue; L-FABP gene had a high expression level in liver tissue; LPL gene had a high expression level in the longissimus muscle. In the longissimus muscle, the expression of H-FABP gene was increased with months of age growth increase, and the highest expression level in24months; the expression of L-FABP gene had a irregularity with months of age growth increase; the expression of LPL gene was increased with months of age growth increase at the beginning, and then enter a plateau, after12months of age, expression no longer increased.The Tianfu goat longissimus muscle IMF content was growth with the increased with months of age. Gene expression level and IMF content correlation analysis showed that:the expression of H--FABP gene and the IMF content showed a highly significant positive correlation, and the correlation coefficient was0.938; the expression of LPL gene and the IMF content was significant with a positive correlation, and the correlation coefficient was0.598; the expression of LPL gene and the IMF content was uncorrelated.
