Influences Of Huajiao-6Decoction On Immunologic Functions In Mice

Objective:The aims of this research is to investigate the influences of Huajiao-6Decoction which is a kind of Mongolian prescription (short for huajiao-6Decoction) on immunologic functions in normal and immunocompromised mice.Methods:The models of non-specific, humoral and cellular immunosuppressed functions were copied by hydrocortisone (HC), cyclophosphamide (CTX) and cyclosporin A (CsA) respectively in mice. Carbon clearance method was used to determine the mononuclear macrophage function, the gravimetric method was used to measure the thymus and spleen weight, the ELISA reagent box was applyed to assay the lysozyme levels in serum and the quantity of the IL-12secreted by macrophages, CCK-8evaluated the activity of the spleen NK cell, to observe the non-specific Immunologic Functions of the huajiao-6Decoction in normal and immunocompromised mice. The hemolysin HClgM and antibody levels (IgQ IgM) of serum were assayed by photometric method and ELISA respectively, so that humoral immunologic functions of the huajiao-6Decoction were observed in normal and immunocompromised mice. The ear conch swelling was determined by Delayed Type Hypersensitivity (DTH) method, and the T Lymphocyte Subsets in peripheral blood was assayed by Flow Cytometry, so that the cellular immunologic functions of the huajiao-6Decoction was observed in normal and immunocompromised mice.Results:Huajiao-6Decoction could significantly improve the mononuclear macrophage functions, organ weight coefficients, lysozyme levels in serum and the activity of the spleen NK cell in normal and HC-induced non-specific immunocompromised mice; In contrast, huajiao-6Decoction significantly inhibited the antibody levels in serum in normal mice and enhanced the inhibitory action by CTX on the humoral immunocompromised mice; huajiao-6Decoction also could significantly inhibit the DTH in normal mice and enhance the inhibitory action by CsA on the cellular immunocompromised mice, Moreover, huajiao-6Decoction could significantly increase the CD3+levels, but significantly modulated CD4+CD8+and CD4+/CD8+levels in normal and CsA-induced cellular immunocompromised mice.Conclusion:Huajiao-6Decoction could significantly improve immune function of normal and HC-induced non-specific immunocompromised mice; and could strikly inhibit the immune function of normal and CTX-induced humoral immunocompromised mice, and modulated the immune function of normal and CsA-induced cellula immunocompromised mice.
