The studies were carried out in the Scientific and Educational Park,Laboratory of Crop Developmental Biology of Henan Agricultural University in2009~2011. The pot, field and indoor cultivation way were used to get the root ofwheat.The study was carried from six aspects as follows: determining the wheat rootactivity circadian changes and optimal sampling time; the effect of wheat root samplestorage time on root activity and the optimum measuring time; similarities anddifferences of wheat roots in pot and field cultivation; the effect of different culturemedia on wheat physiology and root physiological activity; the effects of directflushing root and mesh wrapped irrigation on the measurement of root weight (dryweight); the effects of low temperature treatment on root physiological activity. Sincethe research purpose were different in each test, different methods and techniqueswere used.The main results of this study are as follows:1Determined the wheat root vigor varied significantly within a day: themaximum determination values of root vigor were presented from10:00to14:00, andthereafter the vigor declined until the minimum values were presented from23:00to02:00while from02:00to10:00the vigor change curve roughly took an N shape. Sothe optimal sampling time within a day for field root vigor determination is between10:00and14:00.2Determined the effects of preservation time on the wheat root activity: The rootvigor remained steady within6hours after sampled, then the vigor changed obviouslyespecially after16hours. The results showed that wheat root activity should be testedimmediately after sampled, if the actual circumstances don’t permit, yet thedetermination should be completed within6hours after sampled.3Displayed the difference of wheat root activity between pot and fieldcultivation: When the other conditions were same, there were apparently difference ofwheat root activity between pot and field cultivation, and the potted was stronger than the field. In view of there were great difference in difference culture way, so it shouldbe fully considered in the evaluation of wheat root activity in different treatment.4Elucidate the effects of direct flushing and mesh wrapped irrigation onmeasurement accuracy of root weight (dry weight): Results showed that, thereserious impact on wheat root weight (dry weight) measurement accuracy in differentwashing methods, after get the root-soil samples. The single root weight differencereached significant level between direct flushing root and mesh wrapped root. Theresults show, root weight determination value have significant differences withdifferent washing methods. We recommend the mesh wrapped for the accurate rootweight parameters.5Cleared the influence of low temperature treatment on wheat root activity:The wheat root activity under low temperature treatment show different law(improved TTC method). At0℃and4℃, wheat root activity can keep a relativelylong time. As the storage time prolonged, the root activity slow down. And the rootactivity can keep the longest time stored in4℃. But, the root activity quicklydisappear under-20℃,-40℃,-80℃,-196℃.6Illustrate the effects of different media on wheat seedling plant trait and theroot activity:①Effects on the wheat seedling height: in3~4leaf stage, the effects of differentmedia on the wheat seedling height was not significant, and the lowest is hydroponic.In5~6and6~7leaves stage, the effects of hydroponic and other media on wheatseedling height reached a significant level.②Effects on the weight (fresh weight) of a single plant: in3~4leaf stage, theeffects of different media on weight of a single plant were not significant. In5~6and6~7leaves stage, the difference of weight of a single plant on different media reachedextremely significant level. Combined with the above analysis, in hydroponicconditions, wheat seedling growth had reduced and weight increase trend. Therefore,in indoor culture, should consider to choose the right training mode and media.③The significant effects on wheat root activity: when the other conditions weresame, the order of wheat root activity under different media conditions was:hydroponics>perlite>sand> vermiculite>soil. In view of the effects of different culturemedia on wheat seedling characters and root activity were significantly, the trainingmethods and media should be considered and tagged in the wheat indoor research.
