Study On Rapid Propagation Technique Of Six Bearded IRIS Cultivars

In this research, internode of tillering scape immature of six Bearded Iris varieties is studied with ’Waterloo Bridge’,’White Blue’,’Yellow White’,’White Swan’,’Thrilling’,’Black Flag’ for explants. Research is performed on the tissue culture of Iris establish system of factory production and rapid propagation technology. This text, by different parts of IRIS and different sterilization method, different combinations of plant growth regulators for effect of primary culture, proliferation culture, rooting culture of explants and transplantation groundmass on survival rate etc. With systematic analysis, and in industrial production as a precondition for different the cultivation time on plantlets form, plantlet height, field planting number, illumination type etc. Analysis of physical factors, find the best plantlet growth state as factory production to establish good foundation. The experimental results show that:1. Internode of tillering scape immature which is the best part of Iris, can be directly induced from adventitious buds, while shortening the access time no vaccine. Explants sterilization proposal is:0.1%mercuric chloride sterilizing9min.2. Bearded Iris’Waterloo Bridge’,’White Blue’,’White Swan’the adventitious buds induced from the optimum medium is MS+NAA1.0mg/L+BA3.0mg/L; for’Thriller’ the best medium is MS+NAA1,0mg/L+BA3.Omg/L+2,4-D0.5mg/L; for’Yellow White’and ’Black Flag’ the best medium is MS+NAA0.5mg/L+BA3.Omg/L+2,4-D0.5mg/L.3. Bearded Iris’Waterloo Bridge’,’White Blue’,’Thriller’shoot for optimal proliferation medium is MS+NAA0.5mg/L+6-BA2.Omg/L,’White Swan’ and’Black Flag’shoot for optimal proliferation medium is MS+NAA0.5mg/L+BA1.5mg/L,’Yellow White’ shoot for optimal proliferation culture medium is MS+NAA0.3mg/L+BA1.0mg/L. In the subculture, adventitious bud with bud amount of4-5is best, whose subculture period is30d, shall not exceed6times of subculture.4. Six Bearded Iris species optimum rooting medium is1/2MS+NAA0.5mg/L and three is the best number of rooting plantlets for the strains.5. Six Bearded Iris species for the best transplanting substrate is soil:peat=1:1and six varieties can be without hardening seedling transplanting.6. Six Bearded Iris varieties of factory seedling breeding standards:subculture proliferation culture of standard bottle for seedling is of height3-4cm; every bottle inoculated8plexus; optimum rooting standard bottle for seedling is of height5-6cm; every bottle inoculated10plexus; the best out of the bottle for seedling is of height7-10cm, with root length over3cm. Effective inhibition of vitrification phenomenon of the optimum medium is MS+NAA0.3mg/L+BA1.0mg/L; the best method of prevention of browning in medium supplemented with3g/L is adding activated carbon or1g/L vitamin C. Factory production can also use fountain water instead of distilled water, pH value of which is5.5-6.0.
