With the global salinization of soil area has expanded each year, the production of food crops have been seriously affected. Soybeans as the major economic crops also suffered to cultivate the salinity soybean significance. GmCKR gene which encoded soybean chloroplast ATP synthase F0-Ⅳ subunit is highly expressed in the soybean under the stress70mmol/LNaCl and50mmol/LNaHCO3for24hours.We got the3’end by the3’RACE technology.The whole sequence of GmCKR gene was758bp which contained744bp open reading frame that encoding247amino acids by ORF analysis. GmCKR gene was transferred into Arabidopsis thaliana by Agrobacterium-Mediated.1.We extract the total RNA from leaves and roots in soybeans which under the stress(70mmol/LNaCl and50mmol/LNaHCO3) for24hours, respectively. We got the3’end by the3’RACE technology.The full-length gene was amplified from the cDNA of the leaves. We knew that GmCKR gene was highly conserved sequences by Homologous sequences analysis.2. We got the result that the GmCKR gene can be upregulated under salinity, salt, drought and alkali stress by Realtime PCR method.3. We transformed the pBASTA-CKR recombinant vector into Arabidopsis by Floural Dip method, and we got over-expression transgenic Arabidopsis generations.4. From the phenotypic analysis experiments,we got that the germination rates and growth situations of the transgenic Arabidopsis were much better than wild type. GmCKR gene of over-expression can improve salinity resistance of transgenic plants. In addition, GmCKR genes also has a certain resistance to drought and salt stress.
