With the GM crops released in the global-scale commercialization, the safety evaluation of the GM crops is deepening, and the unintended effects which are beyond the initial goal of phenotypic transformation of GM crops in phenotype, composition, metabolism and so on have become one of the hotspots abroad. We have done research on the agronomic traits, lignin metabolism and other physiological characteristics, and seed nutritional characteristics of generation of T1, T2and T3in transgenic barley transferring LUC and HPT gene, with the original control Gp. It is expected to provide a theoretical basis for safety evaluation.The main results of this report show that the inheritance of LUC and HPT gene in barley varieties tested in the genetic behavior of non-Mendelian modes and both genes were not expressed entirely. Compared with non-transgenic control, the agronomic traits including plant height, leaf area, biomass, ear length, grains per spike, single spike weight and other agronomic traits, stem sclerenchyma, parenchyma, vascular anatomy and other morphological characteristics, chemical composition, content of lignin and the activity of POD and PAL enzyme which are relative to metabolism, chemical composition of leaf and the activity of Nitrogen metabolism-related protein, nitrate reductase, the chemical composition of grain, the content, subunits and protein secondary structure of mineral, albumin, globulin, gliadin and glutenin all show changes in different levels, but the range of changes is different and strains related. Among these, compared with Gp, the Stem mechanical tissue of9T2in transgenic barley were significantly higher; the content of Cu in transgenic barley8T2in the period of heading and maturity were significantly higher; the content of Fe in transgenic barley (8T2,9T2) in the period of maturity was significantly higher; and the brightness and intensity of albumin subunit of39KDa and30KDa around in8T3of transgenic barley showed weaker than those of the Gp ones, while the glutenin subunits of55KDa in8T3were missing. However, due to different technical route, a different donor, recipient and purpose of genes from different sources in the process of genetic modification of plants, the principle called case by case is still necessary. The evaluations on the unintended effects of other transgenic barleys are still needed.
