The Preliminary Research Of The Chemical Constituents From The Stemleaves And The Safety Of Chuanminshen

Chuanmigshen violaceum Sheh et Shan, a traditional Chinese herbal medicine, belongs to Umbelliferae family. It is only distributed in China as Sichuan authentic herbal medicine. Chuanmingshen has the traditional effects of expelling pathogenic wind.clearing heat,invigorating lung and reliecing cough.In clinic, it is frequently applied to treat cough caused by lung-heat,consumption of yin caused by febrile disease and so on. It is widely used in treatment for nourishing Yin both as food and drug.Most of the reports of Chuanmingshen were focused on the chemical components until now. In our early research process,we have made some researches on the quality control methods of the drug,pharmacological effects and the constituents of ethyl acetate extracts from the root of Chuanmingshen.As the Sichuan authentic material, Chuanmingshen serves as health food in many areas, which also has realized industrialization planting.But until now there are not researches about the safety of it.In addition,after the roots of Chuanmingshen were havested,the stemleaves were dropped as wastes by us,which has also caused waste of plant resourses.In order to guarantee the roots of Chuanmingshen can be used safetily and make further development and application of this valuable medicinal resources, we have made preliminary research of the chemical constituents from the stemleaves and the safety of chuanmingshen by modern research methods and techniques.Firstly, we studied on the safety of Chuanmingshen through rat chronic toxicity test.According to investigate the changes of weighK blood routine、blood biochemical、main organ indices and histopatholgy,we have obtained some preliminary results. Secondly, we have made a qualitative and contrastive research on chemical composition between the roots and stemleaves in Chuanminshen by TLC and HPLC.Thirdly,we established the determination method of coumarin from Chuanminshen stemleaves by HPLC with imperatorin as standard substance and made contrastive research on the roots and stemleaves.Fourthly,the determination method of total flavonoids from Chuanminshen stemleaves by UV-Vis spectrophotometry was also established by us.We used rutin as standard substance. At last,we studied on the chemical components from the petroleum ether extracts of Chuanmingshen steamleaves.Three compounds from this extract were isolated in all.The structures of two compounds were identified.They were imperatorin(Ⅰ) and5,8-dimethoxy psoralen(Ⅱ).These results of our study provided a research foundation for guaranteeing safety in clinical practice and promoting further development and application of Chuanmingshen.
