Genome size (C value) is a key biological index. Measuring the C value of a specie not only provides references for genomic sequencing, direct data for evolutionary biology, but also can be useful in ecology and cytobiology.Lithospermum erythrorhizon Sieb. et Zucc is an important medicinal plant, and the root of it contains the active ingredient-shikonin and its derivatives. The study of L. erythrorhizon and shikonin mainly focus on the molecular biology and pharmacochemistry areas, while systemic cognition of its genome remains unclear. Studing L. erythrorhizon on the genome level and genomic sequencing may lay foundations for the regulation of shikonin formation.In present study, we measured the DNA amount of L. erythrorhizon, about1049Mbp, using the flow cytometry by PI colourating, and analyzed its possible biological significance. We also provide some useful information for future studies by comparing different standard plants and buffer solutions.
