| Three cultivar:Favorita, MILA and Chuanyu117were selected as the experiment materials to screening combination of strong sprout and induction of potato in vitro, three experiments, the selection of the best culture medium of vigorous plantlet, the orthogonal combination experiment of induction medium of micro-tubers, and the induction of micro-tubers by adding coumarin or B9into culture medium were used to investigate the best culture medium of vigorous plantlet and the best induction medium of micro-tubers. The results were followed.1The selection of the best culture medium of Strong SproutSucroseã€Activated carbonã€CCC and Uniconazole were used as experimental elements, each element was provided with three horizontal. Five measurement indexes, number of leaf, plant height, number of root,weight and robustness were investigated. The result showed that, A3B2C1D3combination (60g/L Sucrose,1.5g/L activated carbon,50mg/L CCC,0.8mg/L uniconazole) was the best vigorous plantlet combination for Favorita. A2B1C2D3(40g/L sucrose,0.5g/L activated carbon,100mg/L CCC,0.8mg/L uniconazole) was the best vigorous plantlet combination for both Chuanyu117and MILA.2The orthogonal combination experiment of induction medium of micro-tubersCultivarã€Activated carbonã€Coumarinã€B9are experimental elements, each element is provided with three gradients. After60days of culture,count tuber number large tuber numberã€quality of all tubersã€large tuber number rateã€average quality of tuber,and analyze the data.we can conclude that cultivar have the most significant effction on all five indexes, reaches an very significant effect, the sencond is B9.It also have an very significant effection on tuber number large tuber numberã€average quality of tuber〠large tuber number rate; the thrid is activated carbon,it only have an very significant effection on tuber number quality of all tubers; the last is Coumarin,it only have an very significant effection on tuber number,so B9and activated carbon are the ideal induction materials.3Combination experiment of adding Coumarin or B9to induce micro-tuberThe experiment contains seven treatments:one is not add any exogenous plant hormone,three treatments adding with three different gradient of coumarin,and other three treatments adding with three different gradient of B9.After60days harvest micro-tuber. After60days of culture,count tuber number large tuber numberã€quality of all tubersã€large tuber number rateã€average quality of tuber.The results of Favorita as experimental materials shows that when add no exogenous plant hormone and add100mg/L coumarin separately, tuber number of Favorita is the highest. When add100mg/L coumarin separately, large tuber number,quality of all tubers,average quality of tuber are also the highest,the large tuber number rate is higher than other treatments.So,100mg/L coumarin is the ideal concentration for Favorita.The treatment No.five of20mg/L B9is the best concentration for Favorita in all three treatments of adding B9related with tuber number,large tuber number,quality of all tubers,average quality of tuber.The results of Chuanyu117as experimental materials shows that tuber number of treatment not add any exogenous plant hormone is higher than other treatments,but large tuber number large tuber number rate is not ideal. In all three treatments adding with three different levels of coumarin,the No.three treatment of100mg/L coumarin is an ideal concentration for Chuanyul17in all five indexes. In all three treatments adding with three different gradient of B9,80mg/L B9is the best concentration for Chuanyu117.The results of MILA as experimental materials shows that the No.2treatment of50mg/L coumarin is an ideal concentration for MILA in all five indexes of all three treatments adding with three different gradient of coumarin,the average quality of tuber is the heaviest weight.The No.seven treatment of80mg/L B9is an ideal concentration for MILA in all five indexes in all three treatments adding with three different gradient of B9.The tuber number reaches24.67micro-tuber each bottle, large tuber number reaches23micro-tuber each bottle, quality of all tubers reaches4.97g each bottle,so80mg/L B9is the ideal concentration for MILA. |
