28strains of Pleurotus were used as materials in this study. In order to avoid to lose DNA and get good quality DNA of the28Pleurotus strains, we improved the program of extracting DNA called SDS-CTAB. Then, we established and optimized systems of SRAP-PCR and RSAP-PCR on Pleurotus. At last, using UPGMA method to cluster the28strains according to the maps of SRAP and RSAP, respectively, in the hope of finding some new ways to appraise relationship of Pleurotus.After finishing the single factor and orthogonal experiments, SRAP-PCR reaction system and RSAP-PCR reaction system were decided. The SRAP-PCR reaction system was:20ng DNA,2.0μL10×PCR buffer, Mg2+3.0mmol/L, dNTP0.25mmol/L, Primers0.56μmol/L, Taq2.5U, ddH2O13.4μL. The RSAP-PCR reaction system was:30ng DNA,2.0μL10×PCR buffer, Mg2+2.5mmol/L, dNTP0.25mmol/L, Primers0.32μmol/L, Taq2U, ddH2O13.2μL.We used SRAP and RSAP to analyse the relationship of the28Pleurotus strains and genetic polymorphism of them. According to the SRAP,10primers were selected from30primers, which could produce good quality PCR productions. Based on the SRAP analysis,28Pleurotus strains got1566bands totally. In the1566bands contained1562bands which were polymorphic, from50bp to2000bp. The polymorphism rate was99.7%. According to the RSAP,14primers were selected from45primers, which could produce good quality PCR productions. Based on the SRAP analysis,28Pleurotus strains got1604bands totally. In the1604bands contained1603bands which were polymorphic, from50bp to2000bp. The polymorphism rate was99.9%.Using UPGMA method to eatablish clusterings of the28strains according to SRAP maps and RSAP maps, respectively. The results showed that in SRAP-clustering maps, when the similarity coefficient was0.64, they could be classified to4groups. In RSAP-clustering maps, when the similarity coefficient was0.648, they could be classified to5groups.
