The study in this paper by adopting the stem section of the wild hunan Eremochloa ophiuroides as explants in all experiments. The effect of disinfection treatment, different medium, different concentrations of Phytohormones on callus inducing, shoots differentiation and roots inducing were studied.The optimum conditions were found and a full regeneration system of Eremochloa ophiuroides was achieved. The main results of this research were listed as following:1. Disinfection of explants:Selecting the tender stem segments of active growing period as explant. By pretreatment, before puttingin 70% ethanol for 45s. Then the stem segments were surface-disinfected for 10 minutes in HgCl2 (0.1%) with 0.1% Tween-80. Has low browning rate and contamination rate,aboutl2.5% and 6.3% but higher survival rate,about 81.3%.2. Axillary buds induction and increment:The best medium for multiple shoot differentiation is MS medium supplemented with 6-BA 2.0 mg/L, IBA 0.4 mg/L which multiple shoot induction rate is up to 90.0%. MS medium supplemented with 1.0 mg/L 6-BA,0.2 mg/L IBA is the best medium for multiple shoot multiplication culture, and the increment coefficient is up to 3.13.3. Callus induction:The effects of different medium andphytohormones on callus induction were studied which show that MS medium appended with 2 mg/L 2,4-D,0.4 mg/L 6-BA can give highest rate of callus induction, is 64.6%. Which is the most effective subculture medium.4. Callus differentiation:The suggested medium is MS medium supplemented with NAA 0.08 mg/L,6-BA 0.8 mg/L, CH 500 mg/L and 35g/L sucrose can give highest rate of callus differentiation, which is up to 95.0%.5. Roots induction:The suggested medium is 1/2 MS medium supplemented with NAA 0.1 mg/L, IBA 0.5 mg/L,and the roots induction frequency is as high as 100%.6. Acclimatization and transplants:The strong tube-plants with more roots were selected and acclimated in green house for 15 days, before transferred into the hothouses. The surviving rate is up to 80%.
