Study On Chemical Constituents And Determination Of Total Steroids Content From Basidiocarps Of Hericium Coralloides

Hericium coralloides is one of excellent edible anf medicinal macro fungi. Research has proved that fruiting body of Hericium coralloides have Benefit the five internal organs-tonic, aid digestive.This paper studies on chemical constitutes and determination of total steroids content of basidiocarps of Hericium coralloides based on previous studies.Aimed to promote further utilization of this medicinal fungus and provide some basic information for for the comprehensive development of Hericium coralloides.This studies mainly consist of chemical constitutes and determination of total steroids content, the main results as follow:The dried fruiting body of Hericium coralloides were crushed by grinder, then Contintrous reflux extracted by petroleum ether (30-60℃), chloroform, acetone, and methanol.four extracts were obtained. Twelve compounds were isolated from the petroleum ether extracts and chloroform extracts by normal phase silica gel chromatography. Their structures were determined by spectroscopic methods include1H-NMR,13C-NMR,HMQC,HMBC,DEPT and MS. They were identified as9,12-octadecadienoic acid methyl ester (1), ergosta-7,22-dien-3β-ylpalmitate(2), Ergosta-4,6,8(14),22-tetraen-3-one(3), ergosterol peroxide(4), ergosterol(5), Ergosta-7,22-dien-3β-ol(6),Hericenone J(7), cerevisterol(8),ergosta-7,22-dien-3-one(9), β-Sitosterol(10), Hericenes A(11), Hericenes C(12).All of these compounds were separated from the fruiting bodies of Hericium coralloides for the first time.The dried fruiting body of Hericium coralloides were crushed, then continuous reflux extracted by alcohol alkali liquid, the extract liquid recovered solvent by rotary evaporation instrument. then non-saponifiable extracts was obtained by extraction three time with equivalent volume of petroleum ether (30-60℃).Total steriods content was determined by the modified Liebermann-Burchard reagent color, using visible light spectrophotometer. The linear range is0.2-2.4mg, r=0.9998, with precision of RSD were0.51%,0.47%. The repeatability test (RSD) of3.51%. High, medium, low concentrations of recoveries were97.57%,98.69%,96.43%. The stability test (RSD) was2.36%,3.53%. this method the quality of the fruiting bodies of Hericium coralloides total steriods percentage of8.72%.
