9Penniselum ssp. materials from Guangxi Nanning were studied for optimizing the RAPD reaction conditions and procedure. Pennisetum’s genetic diversity and relationship were studied by RAPD markers.The major results showed as follows:1. The obtained optimum scheme was found on reaction system for RAPD-PCR:10×PCR Buffer2.0μL、dNTPs(each10mM)0.4μL、primer(10pM)1.2μL、Mg2+(25mM)1.5μL、TaqE(5U/μL)0.4μL、DNA(10ng)2.0μL、ddH2O up to20μL.2. The analysis of RAPD marker showed that12RAPD primers amplified a total of51scorable markers, percentage of polymorphic loci was100%, Nei’s genetic diversity and shannon’s information index were0.3108and0.4781. The results of RAPD showed that the genetic diversity of forage resources on pennisetum was rich.3. The matrixes of genetic distance and UPGMA cluster were established on RAPD fingerprint.4. The clear and reproducible RAPD bands were recorded according to the electrophoresis results. In the amplification products by the same primer, bands with the same mobility were recorded to a locus,"1"was recorded when the band existed. Otherwise,"0"was recorded. NTSYS software was used for calculation and mapping according to the data. By clustering analysis,9kinds of Pennisetum ssp. can be clearly divided into2categories. Because these9kinds of Pennisetum ssp. can be clearly divided into two distant categories, we can try to use two Pennisetum ssp. from different categories as parent materials for the hybrid breeding. The descendants may have more excellent production traits and phenotypic traits. And then these results may provide scientific basis and theoretical basis for the hybrid breeding of Pennisetum ssp.
