Kenaf, annual herb of Malvaceae Hibscus, is important bast fiber plant,have higher economy value. The genetic diversity of kenaf is abundant.Studies on relationship of kenaf germplasm are important for breeding strategies,provide guidance about parent matching,the extent of genetic variation and the forecast level of heterosis,also important for variety discrimination and widening hereditary basis. Relationship in 60 accessions of kenaf germplasm was assessed using RAPD and ISSR marker.The results are followed:(1)21 primers with polymorphic and clear bands screened from 91 ISSR primers were used to amplify the DNA, 169 bands were amplified based on 21 primers, average 8.05 bands, among which 141 bands were polymorphic, and the percentage of polymorphic bands was 83.4%. The clustering analysis showed: 45 cultivars and 15 wild/half-wild species could be separated at the level of GSC=0.872;45 cultivated kenaf could be divided into four groups at the level of GSC=0.887;The genetic similarity coefficients between cultivars and wild/half-wild species is from 0.450 to 0.970,show higher genetic diversity than 45 cultivars which inner GSC from 0.828 to 0.970. This indicated that there was higher genetic diversity between cultivated kenaf and wild/half-wild species,but lower genetic diversity inner 45 cultivars.(2)38 primers with polymorphic and clear bands screened from 200 RAPD primers were used to amplify the DNA, 286 bands were amplified based on 38 primers, average 7.53 bands, among which 227 bands were polymorphic, and the percentage of polymorphic bands was 79.4%. The clustering analysis showed: 45 cultivars and 15 wild/half-wild species could be separated at the level of GSC=0.870;45 cultivated kenaf could be divided into three groups at the level of GSC=0.872;The genetic similarity coefficients between cultivars and wild/half-wild species is from 0.469 to 0.972,show higher genetic diversity than 45 cultivars which inner GSC from 0.827 to 0.972. This indicated that there was higher genetic diversity between cultivated kenaf and wild/half-wild species,but lower genetic diversity inner 45 cultivars.(3)Combine the data based on RAPD and ISSR,the genetic similarity coefficients among kenaf germplasm were calculated by using NTSYSpc-2.10e software, construct cluster map based on UPGMA cluster analysis, the same results were as follows: 45 cultivars and 15 wild/half-wild species could be separated at the level of GSC=0.871; 45 cultivated kenaf could be divided into four groups at the level of GSC=0.882. The genetic similarity coefficients between cultivars and wild/half-wild species is from 0.457 to 0.971, but GSC from 0.827 to 0.972 inner 45 cultivars.(4) The relationship and genetic diversity of 22 kenaf germplasm resources(from Institue of Bast Fiber Crops,CAAS;Hunan Province)were analyzed by ISSR and RAPD molecular markers, The result of clustering analysis showed:GSC from 0.828 to 0.959 based on ISSR, 22 cultivated kenaf could be divided into four groups at the level of GSC=0.895;GSC from 0.827 to 0.955 based on RAPD, 22 cultivated kenaf could be divided into six groups at the level of GSC=0.898; GSC from 0.836 to 0.954 based on RAPD and ISSR, 22 cultivated kenaf could be divided into five groups at the level of GSC=0.896.(5)RAPD and ISSR methods both could apart 45 cultivars and 15 wild/half-wild species of kenaf, the similar molecular cluster results were obtained from RA PD and ISSR analysis,but ISSR showed higher marker eficiency and diversity detection ability than RAPD molecular method in genetic diference and diversit in kenaf.The study could provide the foundation for Genetic Improvement and Molecular marker-assisted breeding of kenaf..
