Canine distemper virus and Canine parvovirus virus all give rise to worldwide infectious disease and always lead infected dogs to dying. Canine distemper may lead to a variety of clinical symptoms, such as bipolar type fever、acute coryza、bronchitis、 catarrhal pneumonia、gastro-enteritis and neurological symptom. Canine Parvovirus Virus disease’s classical clinical symptoms are enteritis syndrome, puppy often have myocarditis syndrome. People always use specificity hyper immune serum and monoclonal antibody in clinical. Egg yolk immunoglobulin (IgY) has a low cost of production and its operation is easy and can produce a large number of antibodies. The Applications of IgY in the dogs’ treatment are currently reported, but the titer is low and the result of the treatment is not ideal. This study attempts to made tissue inactivation vaccines to improve the titer of IgY, and observe the result of clinical treatment.The first experiment:The study use vero cell and F81cell to multiply CDV and CPV, when80percent of the cells manifest CPE, we collect cells and cell culture medium. The second step is using virus we have collected to cause healthy dogs to be ill, when the dogs manifest clinical symptom and die, we segregate virus from the dead dogs’ internal organs. The third step is measuring the virus’ TCID50, using formaldehyde to inactivate virus, then we produce CDV/CPV bigeminy tissue inactivation vaccines using propolis as immunoadjuvants.The second experiment:We inject tissue inactivation vaccines and cell cultured vaccines into laying hens by two different methods, the laying hens were in the laying season. Then we detect antibody’s titer of egg collected from different groups. The experiment was divided into three groups, every group have ten hens. A GROUP:We inject tissue inactivation vaccines into laying hens in the first day、the eighth day、the twentieth-two day and in the thirtieth day. B GROUP:The time when we inject vaccines into laying hens were same with a group, but the diference between A group and B group is that the vaccines we inject into laying hens of B group are all cell cultured inactivation vaccines. C group are blank group, we inject physiological saline. When the last immunization was completed, we collect eggs every three days until the fiftieth day. We use PEG6000to extract IgY from eggs, then use virus neutralization test and Elisa to detect titer of IgY. The result of experiment showed that:the antibody’s titer of A group is higher than B group, the time required to reach the peak by antibody’s titer of A group is shorter than B group. So we can say that using tissue inactivation vaccines to make IgY is better than the traditional methods.The third experiment:At first, we use IgY to make oral capsule preparations. We divide the dogs naturally infected with canine distemper into two groups:test group and control group, every group have ten dogs; we also divide the dogs naturally infected with Canine parvovirus disease into two groups:test group and control group, every group have ten dogs. The two groups were treated in accordance with conventional treatments, the difference is that the test group feed IgY capsule. At last, we calculate two groups’ recovery rate and recovery time. The result showed that:the recovery rate of test group is higher than control group, the recovery time of test group is shorter than control group. The IgY capsule made in our laboratory have good curative effect on Canine distemper and Canine Parvovirus Virus disease.
