Glucagon-like peptide-1(GLP-1) is a peptide hormone from the distal ileum,r-ectum and colon mucosal L cells synthesis and secretion. GLP-1can improve insu lin resistance, low blood sugar and reduce the weight function, making it a new t arget for the treatment of obesity and diabetes. This research project aims to expl ore the effects of glucagon-like peptide-1[GLP-1(7-36)] on the proliferation and differentiation of3T3-L1cells and its related genes expression. In this experiment, GLP-1on the proliferation and differentiation of3T3-L1cells to explore the dose effect and time effect of the proliferation and differentiation and analyzes the GL P-1cycle of3T3-L1cells, using QPCR further study of the GLP-differentiation r elated gene expression in3T3-L1proliferation. The results are as follows:1. GLP-1(10-7M) can inhibit the proliferation of3T3-Llpreadipocytes(P<0.05);2. GLP-1(10-7M)can affect3T3-L1preadipocytes cell cycle (P<0.05);3. GLP-1(10-7M) significant changes in3T3-L1preadipocyte proliferation,e xpression of related genes P13K, P38and PKCξ(P<0.01).4. GLP-1(10"7M) can significantlyimpact the degree of differentiation of3T3-L1cells (P<0.05).5. GLP-1(10-7M) significantly lowered the expression of differentiation-relate dgenesFAS (P<01)but no significant effect on PPARy and C/EBPa gene expre ssion.These results obtained GLP-1(10-7M) can inhibit the proliferation and differentiat ion of3T3-L1preadipocytes, the proliferation of inhibition relate to cell cycle GO/G1phase block, and from lowing the expression of prolifertion-relate gene PI3K, P38and PKCξ provides a GLP-1affect the3T3-L1preadipocytes proliferation may existwa y.
