This study was conducted to explore the effect of all trans retinoic acid (ATRA)on proliferation and differentiation of murine3T3-L1preadipocytes.3T3-L1preadipocytes were cultured and treated with the absence (0mol/L) or presence of10-9、10-8、10-7、10-6and10-5mol/L ATRA. MTS assay was used to detect the effectof ATRA on proliferation of3T3-L1preadipocytes at0,2,4,6,7and8d, respectively.On the12th day of differentiation, Oil red O staining method were applied to observemorphological changes of the3T3-L1cells in differentiating and were eluted withisopropanol to analyze the degree of differentiation. The expressions of genes relatedto differentiation mRNA were detected by Real-time PCR. The results were showedbelow.(1) During the growth of3T3-L1preadipocytes, compared with the control group(0mol/L),10-9mol/L of ATRA had no significant influence on the proliferation of3T3-L1preadipocytes (P>0.05).10-8,10-7,10-6and10-5mol/L of ATRA significantlyinhibited the proliferation of3T3-L1preadipocytes with the time of growth (P<0.01).10-6and10-5mol/L of ATRA could inhibit the proliferation of3T3-L1preadipocytesmore obviously then others.(2) Compared with the control group (0mol/L),10-7,10-6and10-5mol/L ofATRA inhibited3T3-L1preadipocytes to differentiate into mature adipocytes(P<0.01), and10-9and10-8mol/L of ATRA had no significant influence on thedifferentiation of murine3T3-L1preadipocytes (P>0.05).(3) On the12th day of differentiation, The preadipocyte treated with10-9,10-8,10-7,10-6and10-5mol/L of ATRA showed that the expression of PPARγ, C/EBPα,LXRα, FAS, SCD-1and Glut-4mRNA significantly lessened, as compared with thosein the control group (0mol/L), the difference was significant (P<0.01). (4) On the12th day of differentiation, The preadipocyte treated with10-7,10-6and10-5mol/L of ATRA showed that the expression of SREBP-1c mRNAsignificantly increased, as compared with those in the control group (0mol/L), thedifference was significant (P<0.01). The preadipocyte treated with10-8mol/L ofATRA showed that the expression of ACCα mRNA significantly increased, ascompared with those in the control group (0mol/L), the difference was significant(P<0.01). The preadipocyte treated with10-5mol/L of ATRA showed that theexpression of β-catenin mRNA significantly increased, as compared with those in thecontrol group (0mol/L), the difference was significant (P<0.01).
