In ZheJiang province, according to the germplasm resources investigation and collection of Taxus madia, there are eight cultivation bases of Taxus madia, totally about10000acres, breeding seeding40millions strains every year.By optimizing the extraction and measure methods, the taxol and10-DAB contents of Taxus madia in different producing area and collection seasons have been determined; additionally, the elements content of different cultivation soil have been determined. The results showed that the contents of taxol and10-DAB are significantly changed with different producing area. Within the test years, the contents of taxol and10-DAB are further accumulated with the increasing age, and the highest content is in May. This has provided the theory basis for the germplasm’s seasonal purchase and harvest. The soil factor does not related with the contents of taxol and10-DAB. The taxol is only significantly related to the organic substances content and latitude.The cuttage experiment were developed via designing4growing seedlings matrix, the results showed that the last part of sprout branch is the best for cuttage Taxus madia seedings, the admixture soil (1/3crude soil+1/3Hiru stone+1/3humus soil) is appropriate to match with.The2factors and3levels experiments were designed with forestation line form (factor A) and socket standard (factor B). The results showed that the line form0.5m×1.0m, socket standard50cm (opening broad)×40cm (depth)×30cm(bottom broad) is the best for the planting of Taxus madia.The DCR culture medium with adding BA and IBA is good for armpit buds growing in tissue culture of T. madia.The excellent strains were selected according to the growing properties, the excellent parent strains were selected and carried through hybridize combination, to determine the ripening rate. The results suggested that the compatibility of T. madia in intraspecies is higher than interspecies, the different germplasm resources in same species showed higher hybridize degree.
