Six of the most representative Dark tea (Fuzhuan tea, Pu’er tea, Liubao tea, Jinjian tea,Mizhuan tea, Qingzhuan tea) in China are used as materials. The main chemical components are measured, and the effect on the regulation of blood lipid in rat models with hyperlipidemia has been studied too, further more, the differences protein expression between hyperlipidemia model rats and hyperlipidemia model rats feed Fuzhuan tea have been studied.1.Research on the contents of main biochemical composition in Dark teaThe content of caffeine is the highest in the six kind of Dark tea extract, followed by gallic acid and catechins, little free amino acids has been measured. Content of free amino acid in Jianjin tea is3.56%, the rest of other five kinds of Dark tea have a similar content of free amino acid, it’s about1.6%to2.1%. Polyphenol content are quite different from12%to21%, in accordance with the content in descending order were as following:Liubao tea, Fuzhuan tea, Jinjian tea, Pu’er tea, Mizhuan tea, Qingzhuan tea. HPLC was used to detect the content of catechins and gallic acid, the tesults showed that the total content was consistent with total polyphenol.2.Research on Dark tea regulate serum lipids of hyperlipidemic model ratsCompared with the model control group,rats in Fuzhuan tea, Pu’er tea, Liubao tea and Qingzhuan tea of high doses team effectively ruduced weight that induced by high-fat diet. The results aslo showed that, in high doses teams of Fuzhuan tea, Pu’er tea, Liubao tea, Jinjian tea,Mizhuan tea and Qingzhuan tea, both TC and TG in serum and in liver homogenate of rats were decreased remarkably, the liver index was also decreased remarkably, levels of HDL-C and A/G was increased remarkably, levels of LDL-C、 AS、 ALT were slightly decreased but not significant. Pathologic examination on the liver tissue suggested that hyperlipidemia rats treated with Dark tea extracts had relieved symptoms of hepatocyte degeneration and swelling.3.Differences of liver protein expression in hyperlipidemia model rat deal with Dark teaThe liver of hyperlipidemic rats was used as materials, method with good reproducibility of protein extraction and2-DE operation techniques has been established, nice maps of SDS-PAGE were obtained. Differentially expressed proteins were found between two groups by PDQuest software,11upregulated protein spots were found that abundance changes more than1.5times and had good repeatability,21downregulated protein spots were found that abundance changes more than1.5times and had good repeatability.
