Potato (Solairumt uberosum L.) is the fourth largest food crop in the world. China is the largestpotato-producing country, but the average yield is relatively low. Degradation caused by viruses is themain reason for the low yield of potatoes, which causes very serious harm to the world’s potatoproduction. Since the1970s, the extension of tissue culture techniques, a wide range of virus-freepotato production systems and research and application of the microtuber potato have been developed.It makes significantly shortened of the propagation of the potatoes. Minitubers have been a new formfor preservation of germplasm and production of virus-free potatoes followed the virus-free tuberpotatoes. The minitubers materials are difficult to susceptible to pathogenic microorganisms in theenvironment. Minitubers take the advantages on small size, less weight, non-district by seasons, lessvolume for storage, propagation faster than common ways, and high planting survival rate. It producesand transports easier than other germplasm form as the exchange of germplasm resources as seed, it canbe mass-produced all the year, with short-period, large-scale production to meet a large area ofvirus-free potato source requirements. It is considered the ideal method for the research of the formationmechanism of potatoes by Inducing the formation of potato tube in vitro by tissue culture techniques.Potato cultivar ‘Vict’ used in the trail is introduced in the Netherlands. It is an early maturingvariety, with yellow skin and yellow flesh, shallow eyes, round, short growth period, high yield,uniform. It is suitable for export, and it has potential to replace"older white" and "Favorita" varieties.The effects of different media, photoperiod, temperature, sucrose concentration, parts ofmaterals,and activated carbon concentration on the formation of microtubers were studied in this paper.The results are as followed:1. The best medium for the induction of potato microtubers is MS+10%sucrose+0.1%activatedcarbon.2. The photoperiod induces different in tissue culture, the best solution is alternating of dark-light.3. The effects of temperature are different,(17±1)℃is good for the formation and enlargementof the tube.4. The sucrose concentration on microtuber induction affects differs,which10%can get large tubeseasily.5. The materials from different position have different results, it is strong from the base, and gettubes easier and bigger than from the top.6. Activated carbon concentration of0.1%on the tubers can promote the tube formation.
