| The studies were conducted to determine the releasing discipline of the FGA in vitro simulatedsolution by RP-HPLC,Deer’s intravaginal silica gel forelock is made by Institute of Special EconomicAnimals and Plants,the content of flurogestone acetate is30mg,40mg,50mg,choosing the one whoserelease is stable and CIDR imports bolt to induce sika Estrus synchronization comparison test todetermine the effect of estrus synchronization,combined with the B Ultrasonic monitoring the folliculardynamics,detect the change of reproductive hormones in serum and faeces and and their relevance,findthe effective way of the induction of sika deer estrus synchronization,through compare the variation ofthe FGA in vitro and vivo,the change of reproductive hormones and its relationship with follicularwaves,the level of noninvasive group,disscuss the prospects of noninvasive analysis of fecal steroids.Results indicated as follows:(1)Determination the12day’s releasing discipline of the FGA in vitro simulated solution byRP-HPLC and the result showed the releasing tend of the FGA is lowing gradually.The release quantityof50mg group was the least,which was the most appropriate for being used in estrussynchronization.Three groups as a whole showed significant difference.(2)The effect of Estrus synchronization: a total of20deers,FGA group had six deers estrussynchronization48h in average after the withdrawal of intravaginal silica gel forelock(66%), CIDRgroup had eight deers estrus synchronization in average after the withdrawal of CIDR(80%).The rate ofestrus synchronization in FGA group was lower than the CIDR group.(3)Changes in serum reproductive hormones:There was a big peak of the serum E2concentrations in No.63and No.496sika deer sika on eatrousday. The serum E2concentration of the CIDR group in No. S4sika deer kept decreasing aftersettled,serum peak occurred on Day4after settled(113.4pmol/l),and the serum E2concentrations keepdecreasing again before eatrous. There was a big peak of the serum E2concentrations in No.210and S3sika deer on day2after settled.There was no further peak until the estrous.P4concentrations in CIDRincreased sharply after being buried CIDR,and reached a peak on day2after settled,then down to thebasic level. No.496and63sika deer have shown a gradual upward trend after settled.There was a bigpeak on day4after settled then down to the basic level.(4)The average change of the fecal reproductive hormones:The change of FGA group was in thesame pace. No.496and63sika deer showed a gradual down trend after settled,There was a big peak ofthe serum E2concentrations on day4before eatrous,CIDR group fecal E2hormone appears rise-lower–rise-alternate reduce change,the fecal samples P4concentration in the CIDR group of three deers alsoshowed good consistency,it was significantly similar with Serum levels,P4concentration had a sharp riseafter settled,and reached a peak value on Day2after settled,then slowly decreased to the basal level.FGA group was in an up trend after settled,the P4concentration was on peak of the whole process theday2after settled.(5)Correlation of serum and fecal hormone: The CIDR group serum P4and fecal P4was significantly correlated (P=0.0008),r=0.9766.The CIDR group Serum E2and fecal E2was significantly correlated(P=0.0093),r=0.9199.The FGA group serum E2and fecal E2was significantly correlated(P=0.0479),r=0.8155.The FGA group serum P4and fecal P4was not significantly correlated(P=0.3628),r=0.4565.(6)the change of follicle size:CIDR group follicles was inhibited first then appeared thephenomenon of growth,Typical No.S4sika deer:the follicle of left side was first suppressed,appearedgrowth on day before eatrous.FGA group:the follicle diameter of left side in No.63deer decreaseedfrom4.03cm to3.95cm,The growth of the follicle was suppressed,later in a slow growth trend on day4estrous.(7)Correlation of Follicle and the hormone,Endocrine regulation:P4concentrations with follicle change present significant negative correlation(P <0.05),r=-0.8849.E2concentrations with follicle change present significant positive correlation(P<0.05), r=0.8463.(8) hormone levels of noninvasive group is higher than the invasive group,SAS statistical softwaretest:the E2average concentration of the two group is extreme different.(P<0.01).
