In this study, the carbon sources, the types of hormones et al. in the callus induction andproliferation for Thymus mongolicus Ronn.of labiatae Thymus L.are studied. we definethe suitable conditions of callus induction and proliferation in Thymus mongolicus Ronn.Meanwhile, we study the metabolic relationship between the Thyme essential oil contentof the callus and the different carbon sources and also optimize the essential oil extractioncondition of aseptic Thyme. Besides, we study on the polyploid induction of Thymus seedin vitro.1、Callus induction and proliferation of ThymusThe study of callus induction condition of Thymus shows that the MS medium withNAA2.0mg/L and6-BA1.0mg/L is suitable for leaf callus propagating and the MSmedium with2,4-D0.5mg/L and6-BA1.0mg/L and AgNO30.075mg/L is suitable forstem callus propagating. When we use leaves as explent with30g/L sucrose the callusinduction rate is highest, callus grow best.However, while we use stems as explent,glucose as carbon source the callus induction rate is higher than sucrose.2、In research the different carbon sources on the thyme callus in the metabolism of therelationship between oil content, we found that the accumulation of oil content, sucrosebetter than other types of sugar.3、The optimum extraction condition of aseptic Thyme is: With diethyl ether, solid andliquid in1:7extract48h and essential oil extraction amount is the highest.4、Using colchicine induct thyme seed,the result shows that the part of germinatingseeds which are treated by0.1%-0.2%concentration of colchicines can obtain chimericseedlings. When the concentration of colchicines is0.2%and the time is48h, thepolyploid induction rate is the highest, to be57.1%.
