Rice is one of the three major food crops in the world and it also is the most important food crops in China and the world. The issue of rice suffering cold and low temperature always happened in the global, especially in the double seasons rice planting areas. In the early planting rice stage, rice will be suffered cold easily in bud and seedling stage, resulting in lower seed germination rate and rotten seedlings; In the late planting rice stage, rice will be suffered cold damage easily in heading and flowering stage, resulting in decreased seed setting rate. So researching cold tolerance has great significance to rice production.In this study, we selected190families from F2population, derived from744(strong cold tolerance) and Katy (cold sensitive)) randomly, carried out cold tolerance test in rice bud stage (CTBS), and preliminary mapping QTL. Results are as following:1. Cold tolerance test in parent:parent Katy’s seedling rate is0, showed poor cold resistance; parent744is89.3%, showed strong cold tolerance, and great different cold resistance was detected from two parents in bud stage.2. Cold tolerance test in this190families:the results of CTBS was continuous distribution, and close to normal distribution. It told us CTBS is quantitative trait controlled by multi-gene.3. Selecting109SSR markers that have polymorphism between two parents, and using these markers to detect190families for genotype analysis. Using MAPMARKER3.0software to construct linage map that contained twelve chromosomes and covered rice genome about1436.7cM. The average distance between markers is13.18cM.4. Using QTL IciMapping Version3.2software to detect QTL, and7QTL related to CTBS was found and located in chromosome1RM5~RM1231, chromosome2RM263~RM240, chromosome5RM267~RM146, chromosome6RM527~RM3183, chromosome8RM331~RM339, chromosome11RM167~RM4862, RM4862-RM6091intevals respectively. PVE(Phenotype Variation Explain) value is9.32%,9.43%,4.94%,7.57%,12.34%,7.29%,6.29%.
