| Wolfiporia cocos(W. cocos) Ryv.&Gilbn., named poria cocos (Schw) Wolf, is a high basidiomycete which belongs to the Polyporales. Its underground growth, sclerotium, is a traditional Chinese medicine also used in dietary supplements and natural cosmetics. It has beneficial nutritional and medicinal value.Howere,Systematic studies on the genetic characteristic of W. cocos are rare,mating systerm is uncertain,this preventing the development of breeding techniques. in W. cocos. The most widely used technique is strain selection based on single spores or tissue culture, this leads to the phenomenon of homogenization and degradation, which results to large economic losses. Mating system of W. cocos has been studied, including fruit body inducement, observation of ultramicro structure of fruit body, pairing test and fruiting testIn the experiment of fruit body inducement, the fruit body has been obtained by suitable environment longtime, light inducement, and injury inducement, respectively. The result demonstrated that the fruit body of W. cocos could come into being at a high random but a low rate of fruiting by suitable environment longtime. Light inducement owned a quantity of regulation and the rate of fruiting could be raised through appropriate light treatment. Injury inducement could get a high rate of fruiting, which owned great ability of control.The ultramicro structure of fruit body of wild strain L of W. cocos cultured in lab has been observed by light microscope, SEM and TEM. It was conducted that the basidium of fruit body of W. cocos belonged to the type of bare fruit, and the hymenium was on the both sides ofthe trama, being made up ofthe basidial layer. The basidium was on the hole of wall of hymenium, and there was4sporidium on every basidium. The discrepancy existed between the young and mature sporidium, and the latter was4-6μm long and about2μm wide. Where the sporidium grows existed constriction, and all the sporidium grow non-synchronous. What’s more, some of basidium had3stems only.The strain L and52single-spore strains of W. cocos were collected for pairing and fruiting test. The antagonistic tests between single-spore strains and L, the pairing test among single-spore strains, the antagonistic test between mycelium picked from the boundary formed by harmonious single-spore strains’pairing and either L or2parents of single-spore strains have been accomplished. According to the results, B18and B20were selected as the typical trial strains which could match successfully to identify the mating types of rest single-spore strains. The results demonstrated that the single-spore strains of W. cocos had2kinds of mating type factors, and the quantitative proportion of single-spore strains which had the2kinds of mating type factors was1:1by%test. Further more, the single-spore strains and the mycelium picked from boundary formed by single-spore strains’pairing were selected for the fruiting inducement test. The result illustrated the appearance of fruit body cultured by52single-spore strains within3months but the mycelium of single-spore strains’pairing which was thought to match successfully. Above all, the mating type of W. cocos could be ambipolar heterothallism. |
