| Some experimental results show that rhubarb has a broad-spectrum antiviral effect and has bidirectional regulation effect on animal immune function. In livestock and poultry viral disease controlled in livestock and poultry, it has not been reported that especially the individual as the experimental object, directly on the animal individual dosing for antiviral test, but this has important significance for some animal viral disease control. This research project is the experiment of viral infection by three-yellow chicken, with rhubarb ethanol extract as test drug, with NDV IV strain attenuated, the following several aspects of experimental research were did, and some new research achievements were obtained.Three-yellow chickens were attenuated by NDV IV strain, in second to eighth days after experimental inoculation of chickens, collecting the lung, spleen, liver, glandular stomach, rectum, kidney and bone marrow, the virus hemagglutination titer of various samples were detected with virus hemagglutination test. The test results show, in the collection of the viscera and tissue, in spleen virus content was the highest (at fourth days after inoculation), followed by the lungs and liver (up to fifth days after inoculation). Considering the acquisition and processing of tissue and organ level difficulty, at fourth days after inoculation acquisition of spleen or collected liver at fifth days it is more appropriate to evaluate NDV IV virus content of the experimental chicken. The experiment proved distribution and persistence characteristics of the NDV after inoculation of NDV IV virus attenuated in visceral organ.The experimental conclusion is the basis and premise for antiviral experiment to NDV with rhubarb ethanol extract in chickens in vivo.Antiviral experiment in vivo was did in chicks with rhubarb ethanol extracts of three different dose (1.0mL/kg body weight,time,2.5mL/kg body weight, time and5.0mL/kg body weight, time) by subcutaneous injection and oral administration, in different time periods of the NDV IV strain virus infection(5h-6h before infection,5h-6h post infection,18h-20h post infection), and antiviral effect was evaluated by relative amounts of NDV IV strain virus in the spleen. The test results are that relative contents of NDV IV strain virus in spleen decreased obviously, the chick were treated with three different doses of rhubarb ethanol extract in5h-6h before infection and5h-6h post infection with the NDV IV strain virus; while in18h-20h post infection by NDV IV strain virus, the chick were treated with three different doses of rhubarb ethanol extract, relative contents of NDV IV strain virus in spleen did not decreased obviously. The experimental results show that the prevention has definite curative effect in infection suitable time of the NDV â…£ strain virus with suitable dose of rhubarb ethanol extract, and the curative effect against infection of NDV â…£ strain virus was connected closely with drug dosage and dosing time with rhubarb ethanol extract in experimental chickens, and rhubarb ethanol extract (5.0mL/kg body weight, large dose group) is safe to experimental chicken.This project is the first about anti NDV â…£ virus infection with rhubarb ethanol extracts by chicks in vivo. the conclusion (that is, the prevention has definite curative effect in infection suitable time of the NDV IV strain virus with suitable dose of rhubarb ethanol extract, and the curative effect against infection of NDV â…£ strain virus was connected closely with drug dosage and dosing time with rhubarb ethanol extract in experimental chickens) of this research project also did not see in the literature reports. Therefore the test results has high value and lays a foundation for anti NDV virus and other virus with rhubarb ethanol extract in animal in vivo. |
