Compositions and biological activities of EEP of 29 propolis from 20 provinces of China were researched,in order to know the varieties of compositions and antioxidant activities of propolis from China.It is hoped to base the standardization and industrialization of Chinese propolis.Spectrophotography was used to determine color intensity,E1cm1%,total phenol, total flavonoids,flavone-flavonol and flavanone of EEP.Twenty three standards were selected for HPLC method to confirm the optimum condition.Finger-print was established.Prussian blue spectrophotometry was used to examine total antioxidant activity,and DPPH was utilized to evalue its free radical scavenging activity.Results were as followed.1.Ethanol extractive artwork was optimized.The optimal condition was confirmed according to the yield of total flavonoids.The finally conditions was to extract propolis with 60%ethanol solution under 35℃for 4 hours with solid-liquid ratio 1:20.EEP was obtained by the optimum condition confirmed.And Spectrophotography was used to analyze the physical and chemical characters of EEP. It is showed that different EEP have different chemical characters and compositions. Contents of effect compositions are various in EEP from different areas.EEP from northeast and southwest are similar on yield of EEP,total phenol and total flavonoids. North,middle,east and northwest EEP are similar to each other.EEP from tropic zone are very specific.But the ratio of each composition in EEP are the same, although the content is different.It can be concluded that zone has nothing to do with various compositions of EEP.There are more difference among different color EEP. The lighter color the EEP has,the more effective composition it contains.Yield of EEP is the similar to different propolis with different color.And correlation among E1cm1% total flavonoids and flavone-flavonol is significant,as well as total phenol and flavanone.So,the color maybe the main reason for various compositions of EEP.2.Twenty three standards were chose for separation condition of EEP.Optimum condition of HPLC was made.Reliability of method was examined.It is showed that methanol-0.1%formic acid inhibits trailing of peak effectively as mobile phase. Grouped mixtures can be separate well under 256 and 280 nm.Learner range, correlation,recovery,and limit of detection are all proper to separate and examine EEP.3.Different EEP were detected under the optimum condition.And finger print was established.Although specific peaks were few,it can also reflect common compositions of Chinese propolis based on so many and widely collected samples. Different samples have different combination and content of standards.But p-coumaric acid,cinnamic acid,3,4-dimethoxy cinnamic acid,pinocembrin and chrysin are common of propolis of China.But other standards are not similar to each other on content and combination in different EEP.Especially tropic propolis have few kinds of standards,suggesting special composition.4.Prussian blue spectrophotometry was utilized to examined the total antioxidant activity of EEP.And DPPH was used to determined the free radical scavenging ability.It is showed that different EEP have different bio-activitied.Total antioxidant activity of EEP ranges from 0.44(YN-2) to 2.47(HLJ).AAI value are from 0.46(GD-2) to 9.94(QH).5.Model based on E1cm1%,compositions and antioxidant activities can reflect the relation between compositions and antioxidant activities of EEP.Total antioxidant activity model is Y总抗氧化=0.817+0.004XE1cm1%+0.001X总酚+0.002X总黄酮-0.005X黄酮-黄酮醇 -0.008X黄烷酮;and free radical scavenging ability of DPPH model is YDPPH= 0.385+0.009XE1cm1%+0.005X总酚+0.006X总黄酮-0.017X黄酮-黄酮醇-0.008X黄烷酮.Recently,Chinese propolis are considered low quality product,which is effecting Chinese propolis development.But our results suggest that Chinese propolis have high content of effective compounds and high bio-activities.It is worthy for us to exploit it as functional food and nature antioxidant material.
