Paramisgurnus dabryanus, which belong to Cypriniformes order,Cobitidae family,Cobitinae genus,is widely distributed in China, Japan and Southeast Asia,showing important economic and medicinalvalues. Large quantities of Paramisgurnus dabryanus was exported from China to Japan and Korea everyyear. However, with environment deferioration, Paramisgurnus dabryanus’ production is declining.Resource development and protection become the key to maintain the sustainable development ofParamisgurnus dabryanus.The gene fragments of Paramisgurnus dabryanus’12S rRNA,16S rRNA and CO1are sequenced andanalyzed by three aspects including sequence variation and genetic diversity, genetic structure andphylogeography and population dynamics.It hope to provide a theoretical basis for future resourceprotection and fisheries management.The result showed that:1)398bp for12S rRNA,7hapotypes were detected out of28individualsfrom12S rRNA; The hapotype diversity (HD), mean nucleotide diversity(pi) and average number ofnucleotide differences (K) reached0.442,0.00214and0.849, respectively;2)621bp for16S rRNA,16hapotypes were detected out of32individuals. The hapotype diversity (HD), mean nucleotide diversity(pi)and average number of nucleotide differences (K) reached to0.847,0.00683and3.530, respectively;3)655bp for COI, nucleotide clips to22variation of testing sites,14hapotypes were detected out of28individuals. The hapotype diversity (HD), mean nucleotide diversity(pi) and average number of nucleotide differences (K) reached to0.757,0.0032and2.108, respectively. The genetic diversity detection of16SrRNA and COI is higher than that of12SrRNA.The testing showed that the genetic structure of the Yellow River have no significant differentiation.Fst of12S rRNA is0.1574(p>0.05), gene flow for6.5842, AMOVA test showed that92.36%of thegenetic differences in internal group,7.64%of the genetic differences between populations. For16S rRNA,Fst0.2427(p>0.05), gene flow for20.1516, AMOVA test showed that84.42%of the genetic differencesin internal group,15.58%of the genetic differences between populations. For COI, Fst0.1591(P>0.01),gene flow more than4, AMOVA test showed that91.65%of the genetic differences exist in internal group,and8.35%of the genetic differences existed between populations. It can be concluded that the threeregions of Paramisgurnus dabryanus exchange more fully. Clustering and Network of haplotypes analysisshowed that the three areas have no significant geographical differentiation and enjoy common haplotypes.Paramisgurnus dabryanus, which located in the Yellow River area, whether have populationexpansion needs further research as Tajima ’s D and Fu’ s FS values are not unified.Taken together, the genetic diversity of Paramisgurnus dabryanus is characterized by high haplotypediversity and low nucleotide diversity, more specifically, having genetic divergence, but not achieve thekind of differentiation; existencing a wide range of genes flow but no significant difference of geographyand no population expansion trend. The Paramisgurnus dabryanus lived in Jinan, Shandong Province havehigher genetic variation than that of in Linfen, Shanxi Province and Xinxiang, Henan Province.
