Florfenicol, a kind antibiotic of chloramphenicol, is applied widely. Compared withchloramphenicol and thiamphenico, florfenicol don’t cause aplastic anemia. As a new antibiotic, itis dedicated to the prevention and treatment of animal disease and has showed high antimicrobialactivity. Florfenicol is most effective against microorganisms such as Gram-positive andGram-negative bacteria, mycoplasm, etc. Resistant strains emerge because of improper use offlorfenicol in clinical investigations. Now the existence of resistant genes has been previouslyfound in pasteurella, E. coli, salmonella and staphylococcus. Florfenicol, in a certain dose, hasimmunosuppression effects on rainbow trout and Atlantic cod fish, and reduces Newcastle diseaseantibody titer of the chicken, swine fever antibody titer. In this research, we pay more attention tothe dose scope and using time. The studies on immunosuppression effects of florfenicol in miceprovide us with a basis for subsequent research. Therefore, according to these studies, florfenicolwas applied to establish a necessary theoretical basis for its clinical application. Becauseflorfenicol has immunosuppression effects, we search a kind of immunopotentiator in order toantagonizes the immunosuppression effects of florfenicol. Chinese medicine is less negativeeffects and wide pharmacological functions, so in this experiment we choose bufonis parenteral toinvestigate its immunomodulatory effects on florfenicol-induced immunosuppression in mice.Venenum bufonis, a Chinese herbal medicine, has many pharmacological effects. Our laboratoryhas proved that extract of venenum bufonis, cinobufagin, telocinobufagin, bufonis parenteral, incertain doses, have immunopotentiation effects in mice.In this study we primarily assessed the immunosuppression effects of florfenicol in mice byestablishing OVA-induced immune responses. We next investigated the immunomodulatoryeffects of bufonis parenteral on immunosuppression induced by florfenicol in mice by establishingan animal model of florfenicol-induced immunosuppression. Forfenicol inhibited IgG1, IgG2b,IgG antibody production in serum, the proliferation of spleen cells, T and B lymphocytes.Furthermore forfenicol was shown to have strong inhibitory activity on humoral immunity. Florfenicol significantly inhibited humoral Th2immune responses. The immunosuppressiveeffects of forfenicol were observed in a concentration-dependent manner. That is to say that thehigher of the dose, the stronger of the effects. In contrast, Bufonis parenteral alleviated the spleenatrophy, the inhibition of T and B lymphocytes proliferation, the decline in antibody levels, andthe obstacles to IFN-γ、IL-4、IL-10cytokine secretion induced by florfenicol administration.Bufonis parenteral plays the immunopotentiation effects in a specific range of doses.
