| Looking for a new green feed addition has become a popular research direction becausethe antibiotics brings more and more negative effects as feed addition.Synbiotic, which is anideal substitute to antibiotics, consists of Probiotics and Prebiotics and has the effect ofboth.Lactic acid is widely used as probiotics in many areas like feed, food and so on. However,its cells have the disadvantage of poor tolerance, storage and transportation difficulty, etc. Andlactic acid bacteria spore can just overcome these problems.In sum, this study will optimize thecondition of spore in lactic acid fermentation tank in order to produce a large number ofspores.At the same time, the process for freezing and drying spores should been optimized inorder to improve the survival rate of spores in the process.Testing the spores and other twosynbiotic’s property of heat, acid and bile tolerance so that we can know whether it has a goodtolerability.Finally, the impact on growth and slaughter performance will be observed after weadd the lactic bacteria spores and two kind of synbiotic to the broiler feed.In the experiment, the carbon and nitrogen source has been carefully selected and thefermentation condition has been optimized based on the single factor. Finally, bran turns out tobe the best carbon source and the best nitrogen source is peptone. The spores production rate is83.2%when the conditions are as follows: inoculums size4%, initial pH7.0, incubationtemperature45℃, incubation time72h and stirring speed220r/m.The experiment shows that the highest survival rate of spores is when they are in thelyophilized powder and the trehalose is selected as the protective agent for freeze-dried. Thecondition is that pre-freezing temperature is-70℃and pre-freezing time is4h. In addition, thesurvival rate has been compared under the condition of room temperature and4℃refrigeratorystorage. The result reveals that it makes no obvious difference between two conditions. So itcan be stored under room temperature.The tolerance experiment shows that the lactic bacteria spores and the two synbiotics havesuch a good tolerance of heat, acid and bile tolerance that they could adapt to the feedprocessing and the complex environments in the intestinal of broiler. It indicates that lacticbacteria spores and the two synbiotics are qualified as feed additive. During the whole growth period of broilers(0to42days),We will add a certain percentageof lactic bacteria spores and synbiotics which is composed of lactic acid bacteria spores andtwo other methionine in broiler feed addition. Then we can observe their effect on growth andslaughter performance of broiler.The result shows that adding lactic acid bacteria sporesmenthionine Synbionine in feed addition can effectively promote the growth of broiler andimprove the breast meat rate and positive meat rate in broiler.It turns out that the feed additionwith the liquid methionine and lactic acid bacteria spores is most effective.So the synbioticscomposed of lactic acid bacteria spores and liquid methionine can be used as a green feedadditive to substitute or partly substitute for the use of antibiotics in order to reduce the damageproduced by antibiotic.However, further study about the percentage of spores and methionineand the way and the amount of synbiotic is still needed. |
