The grain glutenine is one of the most important manufacturingproperties of hulless barley. Study of glutenins of hulless barley, especially onthe high-molecular-weight glutenine subunite (HMW-GS), which may be acrucial factor for gluten forming, would provide a scientific foundation forimproving the flour quality and an efficiently using for the flour.123hulless barley varieties and12wild barley accessions were selectedin the study, using a wheat cultivar “Chinese Spring” as the reference. Theglutenine subunits of tested materials were analysed by using the SDS-PAGE,and the HMW-GS genes were also cloned in our study.Our study showed that only three kinds of HMW-GS, which called themA, B and C, were found among135tested materials. For most materials, Aand C type were presented, while the B type appeared in only3materials;besides,8Mexican materials had the B and C type simultaneously. However,the low-molecular-weight glutenine subunite (LWM-GS) were examined asmany as10bands, and4patterns were distinguished according to the generalexpression of bands groups.Meanwhile, we also cloned the genes encoding three HMW-GS, and they were2274bp,2244bp and2124bp in length, encoding757,747and707amino acidssequences, respectively. Sequence analysis showed that the main difference amongthem was the increase or missing numbers of repeated sequences, and the amino acidssequences possessed obvious characters of glutenin proteins.The genetic diversity of HMW-GS in tested materials was unabundantand the variant types were rare. In a word, this study would provide ascientific foundation for improving manufacturing property of hulless barley.
