| In this research, 33 Spring wheat varieties were selected, the Chinese spring wheat was CK, through the SDS-PAGE electricity, the HMW-GS composition and its the variation characteristic has been analyzed. At the same time, Through selected 20 Spring wheat varieties and hybridized cross combinations, we analyzed and determined the HMW-GS of generation and their parents of each hybrid combination. Finally, according to different timing and stages of nitrogen, phosphorus, potassium application, A pot experiment was carried out to study HMW-GS composition and expression. The results were obtained as follows:(1)Glu-A1 encoded four subunits of 1, 1*, 2* and null; Glu-B1 encoded five subunits of 7, 17+18, 6+8, 7+8, 7+9; Glu-D1 encoded two subunits of 2+12, 5+10. There were three major allelic variants at Glu-1 loci in spring wheat varieties in Ning xia, Glu-A1[1(69.70%)], Glu-B1[7(36.36%)], Glu-D1[5+10(84.85%)]. There are 13 HMW-GS types in these spring wheat varieties. The frequency of 1, 7, 5+10 is 69.23%, which is the major type. Good subunit composition is lack. According to the standard grades of Payne, the most varieties Glu-1 quality scores was 6~10, average 8.42, scores points 2.82 in 1A, 1.91 in 1B, 3.69 in 1D. In Payne, the highest score in 1A, 1B, 1D achieved 3, 3 and 4, respectively. The most difference was in 1B. Therefore, we should pay attention to chances the frequency of high quality base of the parent materials in Glu-B1.(2)Through selected 20 spring wheat varieties and hybridized 40 cross combinations, the Chinese spring as CK, we analyzed and determined the HMW-GS of generation and their parents of each hybrid combination. The results showed in most hybrids, the expression of all HMW-GS appeared to be codominant, the band of HMW-GS inherited leaning the female parents. Cytoplasm of female parents was found to have some effect on the expression of very few subunits in a few crosses. At the same time, a new subunit that didn’t exist in the parents was observed in the crosses CB037/09SWH6-2. (3) In the 15kg/acre nitrogen fertilizer levels, HMW-GS of Ningchun NO.42 began at the 5th days after flowering, the 10th days all formed. Subunits formed at different times in different wheat species. There’s effect in compositions of HMW-GS if fertilizer are used, but the fertilizer can effact the final output of HMW-GS. The results showed that the different kinds of fertilizer application can improve the accumulation of HMW-GS. |
