Triptergium wilfordii Hook.f. has been used in clinical treatment of rheumatoid arthritis, lupus erythematosus, chronic kidney disease, psoriasis, and other inflammatory and autoimmune diseases. Sesquiterpene alkaloids component is one of the major active components in Triptergium wilfordii. In this paper, the subject investigated was sesquiterpene alkaloids from Triptergium wilfordii. Combining the modern separation and characterization technique, a systematic method was developed for the extraction, purity and identification of Sesquiterpene alkaloids from Triptergium wilfordii. Finally, different levels of samples were obtained from Triptergium wilfordii including total alkaloid components, coarse fractions, fine fractions and pure compounds.According to the solubility of sesquiterpene alkaloids, Triptergium wilfordii was extracted with ethyl acetate. Total alkaloids were obtained from ethyl acetate extract by using precipitation and solid phase extraction methods. A preparative two-dimensional RPLCX RPLC method was developed to isolate and purify alkaloids from Triptergium wilfordii. And then thirty-one fine fractions were obtained in the first dimension and eleven pure compounds were obtained in the second dimension, all pure compounds were identified by NMR and high resolution MS. The MS fragmentation patterns of the eleven compounds were studied by UPLC/MS/MS and the characteristic fragmentation rules of wilfordate type, evoninate type, iso-evoninate type and hydroxy-wilfordate type alkaloids were obtained under positive ionization mode. Finally,94alkaloids were identified tentatively from Triptergium wilfordii combining the MS fragmentation rules obtained.
