Tripterygium Wilfordii Hook.f is the Tripterygii plant of Celastraceae, which mainly distributes in the southern mountain area of the drainage area of the Changjiang River and the Changbai Mountain in the northeast area. As a kind of traditional Chinese medicine, it is well known for its activities of anti-inflammatory, anti-tumor, and so on. Tripterygium poly-glycoside tablets have been widely used in China for treating a variety of immunologic diseases such as rheumatoid arthritis, lupus erythematosus, nephritis, psoriatic arthritis, and allergic purpura. Based on the production process of Tripterygiuin poly-glycoside pills in Zhejiang DND pharmacy Ltd., Co., the extraction process of Tripterygii extraction and the chromatogram process of Triptolide were studied in the paper.In the paper, the analysis methods of Triptolide and total alkaloids from Tripterygium Wilfordii Hook.f were determined; and the liquid liquid equilibrium data of Triptolide and total alkaloids from Tripterygium Wilfordii Hook.f in the water/chloroform two-phase solvent system were measured. The results show that the extraction yield of Triptolide decreases with temperature, whereas the extraction yields of total alkaloids increases with temperature. However, the ambient temperature is a good choice for the extraction process due to the low boiling point (61.3℃ ) of chloroform, and the process can also meet the industrial demand. In the study of Triptolide chromatogram, the analysis methods of Triptolide from Tripterygium Wilfordii Hook.f. were determined, and the adsorption isotherm was measured. Furthermore, based on the analysis of chromatogram mechanism and industry data, a rate model of column chromatogram was proposed, which could describe the column chromatogram process of Triptolide well. The calculation time of the model is much shorter than industrial operation time. So the model can be used as a soft sensing model, which is of important significance for stabilizing the medicine quality. |
