Obtained from microbial fermentation, erythromycin is a kind of macrolide antibiotics. With a broader antimicrobial spectrum, erythromycin is mainly used in infections caused by penicillin-resistant staphylococcus. In erythromycin industry chain, erythromycin thiocyanate is an important pharmaceutical intermediate. In this paper, macroporous resin is used to refine erythromycin from fermentation broth.The process of separation and purification of erythromycin involved in this paper is as follow:First, use microfiltration membrane to filtrate fermentation broth, and get clear liquor. Second, employ macroporous resin HZ816to adsorb erythromycin from the liquor. After washing resin with alkali buffer solution, desorb erythromycin by butyl acetate. And then, get erythromycin thiocyanate through crystallization. If the relative percentage contents of erythromycin B and C are rich in the fermentation liquour, erythromycin thiocyanate is recrystallized by acetone-water solvent system.1. Fist of all, the analysis method of the process is built in the article. By comparing the yields of titer and erythromycin A, the dates show that the high performance liquid chromatography is more accurate to reflect the process than the determination of erythromycin titer.2. This paper has studied on the separation of erythromycin using maroporous resin. The experimental results demonstrated that maroporous resin can adsorb and concentrate erythromycin fom fermentation liquor effectively.1bed volume resin can process15-20bed volume fermentation liquor with the titer at3000-4000u/ml. what’s more butyl acetate can desorb erythromycin A adsorbed on the resin efficiently, and the amount used is less than2bed volume. The yield of the whole process is more than75%.3. After a series of experiments, it’s found that the quality of the fermentation broth is one of the main factors in the process. If the relative percentage content of erythromycin A in the fermentation liquour is aboveb68%and the the relative percentage content of erythromycinB and C are both less than3%, erythromycin thiocyanate get from the eluent is qualified. And if not, recrystallization is necessary to get high-quality products. The recrystallization process is established in this paper. The content of erythromycin C is reduced by50%to60%through recrystallization.
