Cyclocarya paliurus (Batal.) Iljinskaja (C. paliurus) is a precious plant with a variety of functional roles and active ingredients such as triterpenoids, flavonoids and polysaccharides, and therefore were valuable natural resource for functional food. The extraction conditions for total triterpenoids of C. paliurus leaves was established and optimized, and five triterpenic were separated and identified in this paper. At the same time, antibacterial activities of triterpenoids from C. paliurus leaves was studied.Major factors about extraetion ratio of total triterpenoids had been researched by seleeting alcohol as solution and taking method of ultrasonic-assisted extraction to withdraw. The best extraetion industrial was established by the researched of orthogonal design which was on basis of single factor experiment, and experiments of extraction and extraction times of BuOH. As a result, when alcohol solution65%. solid liquid Proportio1:12and time30, the extraction were optimal parameters, and the extraction ratio of total triterpenoids under the optimal parameters was9.19%. The extraction ratio of2extraction times was94.4%. The reasonable time of extraction by BuOH was two time,and the recoverr ratio of extraction was91.9%.Purity of triterpenoids was purified and discolorated by macro-porous resin by active-ted carbon Through silica gel column chromatography, Sephadex LH-20column chromat-ography and semipreparative high performance liquid chromatography, Five triterpenic were separated from purity of triterpenoids and identified as ursolic acid(UA)、oleanolic acid(OA)、betulinic acid(BA)、2a-hydroxyursolic acid(CA)、2a-hydroxyoleanolic acid(MA) respectively by determination of TLC, HPLC, RRLC-TOF/MS, DEPT,1H NMR and13C NMR. And betulinic acid and2a-hydroxyoleanolic acid were separated and identified from leaves of C. paliurus for the first time.The results of vitro antibacterial activity showed that the total triterpenoids from Cyclocarya paliums leaves had strong anti-bacterial activity against three types of bacteria, and the anti-bacterial activity against Escherichia coli and Staphylococcus aureus was better.When the consisentration of total triterpenoids was24mg/mL,the inhibition zone were32.2±1.08mm and30.6±1.14mm respectively. UA, OA, BA, MA and CA from total triterpenoids had different level of anti-bacterial activity against Staphylococcus aureus and Bacillus subtil is, but had no anti-bacterial activity against Escherichia coli.When consisentrations of MA.CA,OA and UA were3.0mg/mL,the inhibition zones were all bigger than20.0mm.
