| Diabetes mellitus has the characteristics of high blood sugar,which is a serious hazard to human health,and its occurrence is lifelong,and can also cause a series of chronic or acute complications.Therefore,it is very important to find a suitable treatment.At present,there are a variety of hypoglycemic drugs used in clinical treatment,however,most of them have large side effects and drug resistance on the human body,which is not conducive to long-term use.Low-toxicity natural medicines play a unique advantage in the treatment of diabetes.Its efficacy is a comprehensive manifestation of synergy of multiple ingredients and multiple mechanisms.It is not easy to be tolerated,and it is also good for syndromes caused by diabetes.The therapeutic effect of natural medicines,so looking for anti-diabetic drugs from natural medicines has aroused widespread concern.In this paper,two natural products of Cyclocarya paliurus(Batal.)Iljinska and Lithocarpus polystachyus(Wall.)Rehd,which have the same hypoglycemic effect,were used as the research objects.Different polar solvents were used to extract the active parts,and tried to explore the best extraction process.Using distilled water to extract the active components of Cyclocarya paliurus(Batal.)Iljinska,the yield was 26.1%,and using 60%ethanol to extract the active components of polygonatum,the yield can reach24.4%.The main components of the extract were identified by UPLC-MS,and the chemical identification was then carried out based on the high-resolution molecular weight and fragment ion,as well as mass spectral database searching.Finally,313compounds were analyzed from the water extract of Cyclocarya paliurus(Batal.)Iljinska 46 compounds were identified,while 219 compounds were analyzed from the alcohol extract of Lithocarpus polystachyus(Wall.)Rehd and 49 compounds were identified.The inhibitory activity of Lithocarpus polystachyus(Wall.)Rehd extract was evaluated by establishing enzyme inhibitor model.Four flavonoidα-glucosidase inhibitors with good activity were first found from Lithocarpus polystachyus(Wall.)extract were screened by UF-UPLC-MS and identified:naringenin(IC50=10.03±3.01μg/m L),astilbin(IC50=6.14±1.21μg/m L),morin(IC50=8.46±2.16μg/m L).The theoretical prediction of the binding mode of the inhibitor andα-glucosidase was carried out by computer simulation,and the results showed that the stronger the inhibitory activity of the enzyme,the more stable the spatial binding,which explained the result of the activity determination.The total extracts of Cyclocarya paliurus(Batal.)Iljinska and Lithocarpus polystachyus(Wall.)Rehd leaves prepared by the best extraction process were tested for acute toxicity and biological activity in mice.It was found that the two extracts were low toxicity and had biological safety.The hypoglycemic effects of the two extracts were tested in diabetic mice according to different proportions.The hypoglycemic effects of the two extracts were analyzed by observing the weight and blood glucose changes of mice,and the hypoglycemic effects of the two extracts were evaluated by the mouse model.It was found that the ratio of Cyclocarya paliurus(Batal.)Iljinska and Lithocarpus polystachyus(Wall.)Rehd was 2∶1 high dose group【1.2 g/(kg·d)】,which could repair the male reproductive system caused by diabetes.Using the optimal ratio of Cyclocarya paliurus(Batal.)Iljinska and Lithocarpus polystachyus(Wall.)Rehd as the main material,adding other accessories to make hypoglycemic tablets,and conduct pharmacological experiments on the preparation to further verify the safety and hypoglycemic effect of the drug. |
