The study took special mushroom cultivation techniques, research edible fungus degradation of wheat straw generation pleurotus ostreatus polysaccharides(POP)metabolic regularity. Establish a set of technology of separation, extract and detecte POP content and preparation of edible fungi POP from mycoplasm for safe and effective low-cost POP providing the theoretic basis and the application guidance.Series I:Studies on the fungi foster of wheat straw on specified conditionsThis series explore the appropriate cultivation management conditions of pleurotus ostreatus hyphae biodegradate wheat straw.Our experiment data shown that the proper temperature of oyster mushroom hypha grown period is26-28℃,while the oyster mushroom sporophore grown period is12~20℃.The relative air humidity of the oyster mushroom hypha grown period and the oyster mushroom sporophore grown period is65%~70%and85%~95%separately. The shine is an inhibitory factor for hypha grown but some little spread light is useful for sporophore grown. The proper PH of hypha grown is5.5~6.5,we using alkalescent culture medium in our experiments because acid can be produced during the oyster mushroom grown. The sample is collected respectively in nutrition stage, mulberry stage and coral stage as the experiment material for the next reseach.Series II:Studies on the technological process of separation, purification, dynamically detection POP and metabolism of Mushroom fungus degradation wheat stalksBy the orthogonal experimental design,researched the different levels of different factors(Crushing time.soaked time, boiled time and the pH value of treated liquor)on extracting POP to find the better way.The results showed that:fungus substance samples were triturated with26000r/min lmin,soaked48h with acidic treated liquor(pH=3.50) and boiled1h, extraction, repeated twice, the extraction liquid transform into PNMP by centrifuging, inspissating, alcohol precipitation, centrifugation and constant volume.Detecte POP content respectively from mycoplasm by authrone-sulfuric acid method. The results showed that:the two groups POP contents are both show trend of rose at first, then decreased,and reach a peak in First mushroom, they are80.880mg/g,59.048mg/g.Series Ⅲ:Studies on the influence of Absorption metabolic function of fungi source POP on rabbit small intestineEight adult otter rabbits selected were divided into two groups and carried out in vivo experiments(intestinal vascular capsule and jugular vein fistula).60ml perfusion fluid were ingested into small intestine of otter rabbit respectively. the control group composed of perfusion fluid:0.9g methionine,3g glucose, normal saline; the experimental group composed of perfusion fluid:0.9g methionine,3g glucose mycoplasm extract. In order to understand the influence of POP on glucose and methionine metabolism of rabbit intestinal epithelial cells. The result showed that,in the test group, dynamic changes of glucose disappearance rate in the intestinal perfusion fluid at20~160min were all significantly raised (p<0.05); the average rate of serum glucose absorption was also significantly increased, and the serum insulin levels at20~100min were isochronous significantly increased (p<0.05); Met dynamic disappearance rate in the intestinal perfusion fluid for40-100min were all significantly lower (p<0.05); the serum concentration of Met at20-80min increased (p<0.05).these results all showed that POP can effectively promote the glucose absorption of intestinal epithelial cell while the blood glucose level was stable by the help of Insulin and Glu, it can also effectively promote the Met absorption of intestinal epithelial cell,in order to effectively promote the body growth and metabolism.
