The Component Analysis And Bioactivity In Vitro Of Methanol Extracts From Six Kinds Of Edible-Medicinal Fungus

Abstract/Summary:
Edible-medicinal fungus has highly edible and medical value.It has the functions of antioxidation,anti-tumor,enhancing immunity and preventing ’three highs’,and it is the research hotspot of healthy food.For the development of edible-medicinal fungus resources in China,six kinds of edible fungi were selected as the objects in this study,and the effective components was analyzed by GC-MS.The antioxidation,inhibition of α-glucosidase,binding rate of cholate,scavenge of nitrite in vitro and correlational analysis were studied.The main research results are as follows:(1)The yield and the content of efficacy component of six kinds of edible-medicinal fungus were determined.The results have shown that the yield of Phellinus linteus,Inonotus obliquus,Ganodorma lucidum,Hericium erinaceus,Boletus edulis,Oudemansiella radicata were 3.78%,4.08%、1.91%、3.76%、3.85%,4.11%.Total phenols contents were 7.026,1.679,0.867,1.445,16.910,2.062 mg/g.Total flavonoids contents were 18.920,5.185,0.783,0.958,4.113,1.648 mg/g.Ergosterol contents were 37.072,45.780,45.157,0.174,8.213,0.227 μg/g.Adenosine contents were 0.391,0.100,0.283,0.093,1.115,0.040 mg/g.The results showed that the highest yield was obtained from Oudemansiella radicata.The content of total phenol and adenosine in the extract of Boletus edulis was the highest.The content of total flavonoids in the extract of Phellinus linteus was the highest.Ergosterol was the highest in the extracts of Inonotus obliquus.(2)GC-MS was used to analyze the components of extracts from six kinds of ediblemedicinal fungus.The results have shown that it is a large number of elution peaks of Hericium erinaceus,Boletus edulis and Oudemansiella radicata.High content of xylitol and organic acids in Hericium erinaceus.The Boletus edulis contains sugars,alcohols,esters,etc.The organic acids in Oudemansiella radicata were rich and the content was high.But Phellinus linteus,Inonotus obliquus and Ganodorma lucidum have a few peaks,and a small amount of sugars and unsaturated fatty acid were detected.(3)The antioxidative effects of extracts from six kinds of edible-medicinal fungus in vitro were analyzed and compared.The results have shown that Phellinus linteus has the highest content of DPPH·scavenging,ABTS·scavenging,relative reducing power and anti-lipid peroxidation power.Scavenging activities of hydroxyl free radicals of Oudemansiella radicata were higher than other edible-medicinal fungus.(4)The biological activity of extracts from six kinds of edible-medicinal fungus in vitro were analyzed and compared.The results have shown that Phellinus linteus and Inonotus obliquus showed the best inhibition of α-glucosidase,deffer from Ganodorma lucidum or Boletus edulis.The IC50 values range from 0.053 to 16.623 mg/mL.There was a little difference of binding rate of cholate in six kinds of edible-medicinal fungus.Ganodorma lucidum has the best binding capacity of cholate,followed by Boletus edulis.Oudemansiella radicata showed the best scavenge of nitrite,which was significantly higher than other edible-medicinal fungus.(5)Correlational analysis between efficacy component and activity were conducted.The DPPH·scavenging,ABTS·scavenging,relative reducing power and anti-lipid peroxidation power were relating intensely with total flavonoids and ergosterol contents.The anti-lipid peroxidation was relating intensely with total phenols contents.Adenosine contents was not correlated with antioxidant indexes.Antioxidant indexes were more affected from total phenols,total flavonoids and ergosterol than adenosine.The correlation in vitro activity showed that,the inhibition of αglucosidase and scavenge of nitrite were relating intensely with total phenols and adenosine contents,but weakly with total flavonoids and ergosterol contents.There was no correlation betw een four components and binding capacity of.cholate.The main components detected in the extracts of six kinds of edible-medicinal fungus by GC-MS were organic acids,sugars,alcohols,esters,amino acids,etc.These components have been proved to have antioxidation and hypolipidemic effect.So in this study,the antioxidation,inhibition of α-glucosidase and binding capacity of cholate of six kinds of edible-medicinal fungus may intensely with these components.Organic acids,sugars,alcohols,esters,amino acids,etc,may were relating intensely with the flavor characteristics of edible-medicinal fungus.In summary,edible-medicinal fungus are rich in nutrients,with different effective components and bioactivity in vitro.This study would provide theoretical reference for screening of edible-medicinal fungus with antioxidant,hypoglycemic and hypolipidemic functions.
